Résumé : Genome-wide association studies (GWAS) have identified more than 100 genetic variants contributing to BMI, a measure of body size, or waist-to-hip ratio (adjusted for BMI, WHRadjBMI), a measure of body shape. Body size and shape change as people grow older and these changes differ substantially between men and women. To systematically screen for age- and/or sex-specific effects of genetic variants on BMI and WHRadjBMI, we performed meta-analyses of 114 studies (up to 320,485 individuals of European descent) with genome-wide chip and/or Metabochip data by the Genetic Investigation of Anthropometric Traits (GIANT) Consortium. Each study tested the association of up to ~2.8M SNPs with BMI and WHRadjBMI in four strata (men ≤50y, men >50y, women ≤50y, women >50y) and summary statistics were combined in stratum-specific meta-analyses. We then screened for variants that showed age-specific effects (G x AGE), sex-specific effects (G x SEX) or age-specific effects that differed between men and women (G x AGE x SEX). For BMI, we identified 15 loci (11 previously established for main effects, four novel) that showed significant (FDR<5%) age-specific effects, of which 11 had larger effects in younger (<50y) than in older adults (≥50y). No sex-dependent effects were identified for BMI. For WHRadjBMI, we identified 44 loci (27 previously established for main effects, 17 novel) with sex-specific effects, of which 28 showed larger effects in women than in men, five showed larger effects in men than in women, and 11 showed opposite effects between sexes. No age-dependent effects were identified for WHRadjBMI. This is the first genome-wide interaction meta-analysis to report convincing evidence of age-dependent genetic effects on BMI. In addition, we confirm the sex-specificity of genetic effects on WHRadjBMI. These results may provide further insights into the biology that underlies weight change with age or the sexually dimorphism of body shape.

Les sources des 240 notices proviennent de la consultation de revues spécialisées et d’archives familiales. Une trentaine de photographies et de caricatures agrémentent l’ensemble. C’est aussi une façon de redécouvrir des sites et des musées régionaux. Nous livrons le témoignage d’écrivains comme Gustave Flaubert, Romain Rolland. Au-delà, nous rendons hommage au Père Lambiez qui, à Bavay, lança en 1790 la première revue archéologique, Feuille d’Or. Un tableau replace les trouvailles du nord de la France dans le contexte des principales découvertes contemporaines (Abou Simbel, Vix, Machu Pichu, Lucy…).

Résumé : 3502 Background: The SIRFLOX study was designed to assess the efficacy and safety of combining FOLFOX chemotherapy (\+/-\ bev) with SIRT using yttrium-90 (Y-90) resin microspheres as first-line treatment of pts with liver metastases from mCRC. Methods: SIRFLOX was an international, multi-center, open-label, RCT in chemotherapy-naive pts with non-resectable, liver only or liver dominant (liver plus lung and/or lymph node metastases) mCRC. Arm A: mFOLFOX6 \+/-\ bev was compared to arm B: mFOLFOX6 + SIRT (SIR-Spheres; Sirtex) administered once with cycle 1 \+/-\ bev until disease progression. The primary endpoint was progression free survival (PFS) using RECIST v1.0. Stratification variables included presence of extra hepatic disease (EHD; liver only v liver dominant), degree of liver involvement ( [≤] 25% v \textgreater 25%), and treatment with bev (at clinician discretion). Results: From Oct 2006 to Apr 2013,530 pts were randomised (arm A, n = 263; arm B, n = 267), 212 (40%) had EHD. Median follow-up was 36.1 months. The median overall PFS was 10.2 v 10.7 months in arms A v B respectively (hazard ratio [HR]: 0.93; 95% CI 0.77-1.12; p=0.428) by Kaplan Meier analysis. The median PFS in the liver was 12.6 v 20.5 months in arm A v B (HR: 0.69; 95% CI 0.55-0.90; p = 0.002) by competing risk analysis. Overall response rate (PR + CR) was 68.0% v 76.4% in arm A v B, respectively (p = 0.113). Hepatic response rate was 68.8% v 78.7% in arm A v B (p = 0.042), including CR rate 1.9% v 6.0% (p = 0.02). The liver resection rate was 13.7% v 14.2% in arm A v B (p = 0.857). Adverse events [≥] grade 3 were noted in 73.3% v 85.4% of pts in arm A v B. Most common toxicities were hematologic; 32.9% v 51.2% and gastrointestinal; 21.2% v 32.9%, including gastric ulcer 0.0% v2.4%. Conclusion: In first-line treatment of pts with non-resectable CRC liver metastases, the addition of SIRT to standard chemotherapy failed to improve overall PFS. However, median liver PFS was significantly extended. The addition of SIRT was associated with acceptable toxicity. Overall survival analyses, combining data from SIRFLOX and two other ongoing studies in this disease setting, are awaited. Clinical trial information: NCT00724503

Résumé : H₂O, CO₂, SO₂, O₂, H₂, H₂S, HCl, chlorinated hydrocarbons, NO, and other trace gases were evolved during pyrolysis of two mudstone samples acquired by the Curiosity rover at Yellowknife Bay within Gale crater, Mars. H₂O/OH-bearing phases included 2:1 phyllosilicate(s), bassanite, akaganeite, and amorphous materials. Thermal decomposition of carbonates and combustion of organic materials are candidate sources for the CO₂. Concurrent evolution of O₂ and chlorinated hydrocarbons suggests the presence of oxychlorine phase(s). Sulfides are likely sources for sulfur-bearing species. Higher abundances of chlorinated hydrocarbons in the mudstone compared with Rocknest windblown materials previously analyzed by Curiosity suggest that indigenous martian or meteoritic organic carbon sources may be preserved in the mudstone; however, the carbon source for the chlorinated hydrocarbons is not definitively of martian origin.

Résumé : It is classically admitted that the goodfriction properties of lamellar compounds are strongly related to their anisotropic structure and especially to the existence of weak interlayer interactions through the van der Waals gap separating the basal layers. As it is also known, the presence of the van der Waals gap in the structure of lamellar compounds will allow lot of chemical species to be intercalated in the structure leading both to the expansion of structure parameters and inter layer interactions modifications. The present work is concerned with the experimental and theoretical study of friction propertiesof Graphite Intercalated Compounds (GICs) in order to better understand thetribologiclamellar compounds. In order to modulate the interlayer interactions, two types of intercalated species were used, electrophylic species (AlCl3, FeCl3, SbCl5) and nucleophilic species (Li, K, Rb).

Résumé : Thiswork examines the relevance of the inclusion of ground-based gravity data in the calibration process of a global rainfall-discharge reservoir model. The analysis is performed for the Durzon karst system (Larzac, France). The first part of the study focuses on the hydrological interpretation of the ground-based gravity measurements. The second part of the study investigates further the information content of the gravity data with respect to water storage dynamics modelling.The gravity-derived information is found unable to either reduce equifinality of the single-objective, dischargebased model calibration process or enhance model performance through assimilation.

Résumé : Elevated resting heart rate is associated with greater risk of cardiovascular disease and mortality. In a 2-stage meta-analysis of genome-wide association studies in up to 181,171 individuals, we identified 14 new loci associated with heart rate and confirmed associations with all 7 previously established loci. Experimental downregulation of gene expression in Drosophila melanogaster and Danio rerio identified 20 genes at 11 loci that are relevant for heart rate regulation and highlight a role for genes involved in signal transmission, embryonic cardiac development and the pathophysiology of dilated cardiomyopathy, congenital heart failure and/or sudden cardiac death. In addition, genetic susceptibility to increased heart rate is associated with altered cardiac conduction and reduced risk of sick sinus syndrome, and both heart rate–increasing and heart rate–decreasing variants associate with risk of atrial fibrillation. Our findings provide fresh insights into the mechanisms regulating heart rate and identify new therapeutic targets.

Résumé : Samples from the Rocknest aeolian deposit were heated to ~835°C under helium flow and evolved gases analyzed by Curiosity's Sample Analysis at Mars instrument suite. H2O, SO2, CO2, and O2 were the major gases released. Water abundance (1.5 to 3 weight percent) and release temperature suggest that H2O is bound within an amorphous component of the sample. Decomposition of fine-grained Fe or Mg carbonate is the likely source of much of the evolved CO2. Evolved O2 is coincident with the release of Cl, suggesting that oxygen is produced from thermal decomposition of an oxychloride compound. Elevated δD values are consistent with recent atmospheric exchange. Carbon isotopes indicate multiple carbon sources in the fines. Several simple organic compounds were detected, but they are not definitively martian in origin.
