Résumé : Peroxidases have been shown to be involved in the polymerization of lignin precursors, but it remains unclear whether laccases (EC 1.10.3.2) participate in constitutive lignification. We addressed this issue by studying laccase T-DNA insertion mutants in Arabidopsis thaliana. We identified two genes, LAC4 and LAC17, which are strongly expressed in stems. LAC17 was mainly expressed in the interfascicular fibers, whereas LAC4 was expressed in vascular bundles and interfascicular fibers. We produced two double mutants by crossing the LAC17 (lac17) mutant with two LAC4 mutants (lac4-1 and lac4-2). The single and double mutants grew normally in greenhouse conditions. The single mutants had moderately low lignin levels, whereas the stems of lac4-1 lac17 and lac4-2 lac17 mutants had lignin contents that were 20 and 40% lower than those of the control, respectively. These lower lignin levels resulted in higher saccharification yields. Thioacidolysis revealed that disrupting LAC17 principally affected the deposition of G lignin units in the interfascicular fibers and that complementation of lac17 with LAC17 restored a normal lignin profile. This study provides evidence that both LAC4 and LAC17 contribute to the constitutive lignification of Arabidopsis stems and that LAC17 is involved in the deposition of G lignin units in fibers.

Résumé : The subjects presented here are very different. Their common feature is that they all involve quantum phenomena in a gravitational field: gravitational quantum states of ultracold antihydrogen above a material surface and measuring a gravitational interaction of antihydrogen in AEGIS, a quantum trampoline for ultracold atoms, and a hypothesis on naturally occurring gravitational quantum states, an Eötvös-type experiment with cold neutrons and others. Considering them together, however, we could learn that they have many common points both in physics and in methodology.

Résumé : A life-long follow-up of physiological and behavioural functions was initiated in 38-month-old mouse lemurs (Microcebus murinus) to test whether caloric restriction (CR) or a potential mimetic compound, resveratrol (RSV), can delay the ageing process and the onset of age-related diseases. Based on their potential survival of 12 years, mouse lemurs were assigned to three different groups: a control (CTL) group fed ad libitum, a CR group fed 70% of the CTL caloric intake and a RSV group (200 mg/kg.day(-1)) fed ad libitum. Since this prosimian primate exhibits a marked annual rhythm in body mass gain during winter, animals were tested throughout the year to assess body composition, daily energy expenditure (DEE), resting metabolic rate (RMR), physical activity and hormonal levels. After 1 year, all mouse lemurs seemed in good health. CR animals showed a significantly decreased body mass compared with the other groups during long day period only. CR or RSV treatments did not affect body composition. CR induced a decrease in DEE without changes in RMR, whereas RSV induced a concomitant increase in DEE and RMR without any obvious modification of locomotor activity in both groups. Hormonal levels remained similar in each group. In summary, after 1 year of treatment CR and RSV induced differential metabolic responses but animals successfully acclimated to their imposed diets. The RESTRIKAL study can now be safely undertaken on a long-term basis to determine whether age-associated alterations in mouse lemurs are delayed with CR and if RSV can mimic these effects.

Résumé : Rats introduced on islands can affect ecosystem structure and function by feeding on terrestrial plants and both marine and terrestrial animals. The diet and trophic position of Rattus rattus introduced on Ile de la Possession (Iles Crozet) was assessed from two sites, according to the presence or absence of a king penguin colony. We used three complementary assays: macroanalyses of the stomach, faecal microhistology, and stable isotope analyses of d15N/d13C in liver. Near the rookery, spermatophytes contributed on average 50% (confidential interval: 23-75) to the diet based on isotopes, mainly consisting in reproductive parts of Poa spp., Agrostis magellanica, and Cerastium fontanum identified in faeces. Terrestrial animal preys were represented by insects that contributed 25% (0-56) in isotopes, dominated in faeces by caterpillars of Pringleophaga spp. and adult weevils. Bird remains were found in faeces, forming 18% (6-30) of isotopes. Terrestrial earthworms contributed to 7% (0-21), with chaetae observed in faeces. On the other site, spermatophytes represented 62% (51-73) of assimilated food in rats' livers, mainly formed by Poaceae and Acaena magellanica, insects by caterpillars [24% (10-39)], and terrestrial earthworms [13% (2-23)]. Our results suggest that rats, which were found at the top of terrestrial food chains, may have a direct role on a such simplified ecosystem, by preying on the most abundant and largest body-sized terrestrial invertebrates,--e.g. the keystone species Pringleophaga spp.--, and by feeding on both reproductive and vegetative parts of autochthonous and introduced plants. The discrepancies and usefulness of employing both isotopes and faecal analyses are discussed.

Résumé : This paper presents the work done during the first year of the EPIS project. This project deals with the process of conductingmultiple and parallelmulti agents-based simulations (MABS) on a cluster or a grid in order to generate sufficient data for scientific use (e.g. in the case of a sensibility analysis of a simulation). We provide a new, general and user-friendly approach to marry MABS and High- Performance Computing (HPC). We, thus, propose a workflow and an associated HPC infrastructure. These two permit to easily deploy a lot of simulations on a cluster without any prior parallelizing work. The method wants to be as generic as possible: no particular MABS targeted, no overhead and HPC compliance work has to be done only once. Moreover the user is guided by a web interface that handles the workflow.

Résumé : Detection of small hepatocellular carcinoma (HCC) eligible for curative treatment is increased by surveillance, but its optimal periodicity is still debated. Thus, this randomized trial compared two ultrasonographic (US) periodicities: 3 months versus 6 months. A multicenter randomized trial was conducted in France and Belgium (43 sites). Patients with histologically proven compensated cirrhosis were randomized into two groups: US every 6 months (Gr6M) or 3 months (Gr3M). For each focal lesion detected, diagnostic procedures were performed according to European Association for the Study of the Liver guidelines. Cumulative incidence of events was estimated, then compared using Gray's test. The prevalence of HCC ≤30 mm in diameter was the main endpoint. A sample size of 1,200 patients was required. A total of 1,278 patients were randomized (Gr3M, n = 640; Gr6M, n = 638; alcohol 39.2%, hepatitis C virus 44.1%, hepatitis B virus 12.5%). At least one focal lesion was detected in 358 patients (28%) but HCC was confirmed in only 123 (9.6%) (uninodular 58.5%, ≤30 mm in diameter 74%). Focal-lesion incidence was not different between Gr3M and Gr6M groups (2-year estimates, 20.4% versus 13.2%, P = 0.067) but incidence of lesions ≤10 mm was increased (41% in Gr3M versus 28% in Gr6M, P = 0.002). No difference in either HCC incidence (P = 0.13) or in prevalence of tumors ≤30 mm in diameter (79% versus 70%, P = 0.30) was observed between the randomized groups. Conclusion: US surveillance, performed every 3 months, detects more small focal lesions than US every 6 months, but does not improve detection of small HCC, probably because of limitations in recall procedures. Résumé : The coupling between a microalgal pond and an anaerobic digester is a promising alternative for sustainable energy production by transforming carbon dioxide into methane using solar energy. In this paper, we demonstrate the ability of the original ADM1 model and a modified version (based on Contois kinetics for the hydrolysis steps) to represent microalgae anaerobic digestion. Simulations were compared to experimental data of an anaerobic digester fed with Chlorella vulgaris. The modified ADM1 fits adequately the data for the considered 140 day experiment encompassing a variety of influent load and flow rates. It turns out to be a reliable predictive tool for optimising the coupling of microalgae with anaerobic digestion processes.
