Résumé : Remorins (REMs) are proteins of unknown function specific to vascular plants. We have used imaging and biochemical approaches and in situ labeling to demonstrate that REM clusters at plasmodesmata and in approximately 70-nm membrane domains, similar to lipid rafts, in the cytosolic leaflet of the plasma membrane. From a manipulation of REM levels in transgenic tomato (Solanum lycopersicum) plants, we show that Potato virus X (PVX) movement is inversely related to REM accumulation. We show that REM can interact physically with the movement protein TRIPLE GENE BLOCK PROTEIN1 from PVX. Based on the localization of REM and its impact on virus macromolecular trafficking, we discuss the potential for lipid rafts to act as functional components in plasmodesmata and the plasma membrane.

Résumé : The hydrolysis and condensation of a silylated derivative of ureidopyrimidinone led to nanostructured hybrid silica, such as that depicted, as clearly shown by powder XRD studies. The nanostructuring was directly related to molecular recognition through hydrogen bonding. By combining FTIR, solution and solid-state NMR spectroscopic data, the transcription of the hydrogen-bonding networks from the precursor to the final product was clearly evidenced.

Résumé : In several neurodegenerative diseases of the CNS, oligodendrocytes are implicated in an inflammatory process associated with altered levels of oxysterols and inflammatory enzymes such as secreted phospholipase A2 (sPLA2). In view of the scarce literature related to this topic, we investigated oxysterol effects on these myelinating glial cells. Natural oxysterol 25-hydroxycholesterol (25-OH; 1 and 10 muM) altered oligodendrocyte cell line (158N) morphology and triggered apoptosis (75% of apoptosis after 72 h). These effects were mimicked by 22(S)-OH (1 and 10 muM) which does not activate liver X receptor (LXR) but not by a synthetic LXR ligand (T0901317). Therefore, oxysterol-induced apoptosis appears to be independent of LXR. Interestingly, sPLA2 type IIA (sPLA2-IIA) over-expression partially rescued 158N cells from oxysterol-induced apoptosis. In fact, 25-OH, 24(S)-OH, and T0901317 stimulated sPLA2-IIA promoter and sPLA2 activity in oligodendrocyte cell line. Accordingly, administration of T0901317 to mice enhanced sPLA2 activity in brain extracts by twofold. Short interfering RNA strategy allowed to establish that stimulation of sPLA2-IIA is mediated by pregnane X receptor (PXR) at high oxysterol concentration (10 muM) and by LXR beta at basal oxysterol concentration. Finally, GC coupled to mass spectrometry established that oligodendrocytes contain oxysterols and express their biosynthetic enzymes, suggesting that they may act through autocrine/paracrine mechanism. Our results show the diversity of oxysterol signalling in the CNS and highlight the positive effects of the LXR/PXR pathway which may open new perspectives in the treatment of demyelinating and neurodegenerative diseases.

Résumé : We previously reported a strong IL4I1 gene expression in primary mediastinal B-cell lymphoma (PMBL) and recently identified the protein as a secreted L-phenylalanine oxidase, physiologically expressed by myeloid cells, which inhibits T-cell proliferation in vitro. Here, we analyzed the pattern of IL4I1 protein expression in 315 human lymphoid and non-lymphoid malignancies. Besides PMBL, IL4I1 expression in tumors was very frequent. IL4I1 was detected in tumor-associated macrophages from most of the tumors and in neoplastic cells from follicular lymphoma, classic and nodular lymphocyte predominant Hodgkin lymphomas and small lymphocytic lymphoma, three of which are germinal center derived. IL4I1-positive tumor cells were also detected in rare cases of solid cancers, mainly mesothelioma. The enzymatic activity paralleled protein expression, suggesting that IL4I1 is functional in vivo. Depending on the tumor type, IL4I1 may impact on different infiltrating lymphocyte populations with consequences on tumor evolution. In the particular case of follicular lymphoma cells, which are susceptible to antitumor cytotoxic T cells killing but depend on interactions with local T helper cells for survival, a high level of IL4I1 expression seems associated with the absence of bone marrow involvement and a better outcome. These findings plead for an evaluation of IL4I1 as a prognosis factor.

Résumé : Uncertainties in model projections of future climate change are high, and have not decreased substantially over the last 30 years: IPCC AR4 range of climate sensitivity - the increase in globally averaged surface temperature expected for a doubling of CO2 - is [1.5 °C, 4.5 °C]. The causes and nature of this uncertainty, more specifically whether or not it will be possible to reduce it, is subject to debate. Before digging into this problem, we start by focusing on a basic, somewhat epistemological question: what is uncertainty in the context of climate science, and how should it be quantified? While this question may at first appear trivial, we claim that it has occasionally proved to be a source of confusion in the aforementioned debate. We illustrate this point by revisiting one among the most significant contributions to this debate in the recent past (Roe and Baker, 2007) and highlight the fact that its conclusions are dramatically affected by the chosen definition of uncertainty. To resolve this issue, we propose widely used standard deviation as a unique, broadly applicable definition of uncertainty. We then recall a general result on the propagation of uncertainty available from Probability theory under this definition, and analyse its implication on a simple stochastic Climate toy model. This analysis suggests that high uncertainty is not an inevitable and general consequence of the nature of the climate system, and hence is not doomed to remain high. More specifically, it suggests that reducing uncertainty on feedbacks in GCMs, through an improved understanding of involved physical processes, does lead to a reduction of uncertainty on climate sensitivity projected by models. Finally, we elaborate on foreseeable advances in Climate research and modeling that may lead to a decrease of feedback uncertainty, and subsequently to a reduction of uncertainty on climate sensitivity in the near future.
