Résumé : Seeds play a fundamental role in colonization of the environment by spermatophytes, and seeds harvested from crops are the main food source for human beings. Knowledge of seed biology is therefore important for both fundamental and applied issues. This review on seed biology illustrates the important progress made in the field of Arabidopsis seed research over the last decade. Access to 'omics' tools, including the inventory of genes deduced from sequencing of the Arabidopsis genome, has speeded up the analysis of biological functions operating in seeds. This review covers the following processes: seed and seed coat development, seed reserve accumulation, seed dormancy and seed germination. We present new insights in these various fields and describe ongoing biotechnology approaches to improve seed characteristics in crops.
have continued to be exploited over the past decade, despite experimental difficulties resulting from gene duplications/redundancies (e.g. identification of the GA receptor), and will obviously continue to be valuable. However, the study of seed biodiversity, an important topic in terms of evolution, cannot be restricted to the analysis of intra-specific natural variation in Arabidopsis. It is time to look at biological diversity in other seedproducing plants in order to understand what makes them unique in terms of biochemical composition, tolerance to adverse environmental conditions, germination features, competition with other plants, and many other processes. Here we review the present status of Arabidopsis seed research and its biotechnological impact. This research will provide a valuable foundation for studies in other species.

Résumé : Neutral and cationic halogen loss of singly and doubly ionised 5-X-uracil (X = F, Cl, Br, I) after collisions with 100 keV protons have been studied in the gas phase. The rates of these dissociation channels are strongly dependant on the nature of the halogen substituent. It is very weak in the case of fluorine but is a dominant channel for iodine. Dissociation mechanisms are proposed for a number of significant channels associated to dehalogenation. It is suggested that some final ion products originate from specific processes. For instance, ion products of mass 38, 39 and 40 amu are very sensitive to the nature of the primarily ejected halogen and result from pathways associated to dehydrogenation and transient formation of the dehalogenated uracil cation.

Résumé : Abstract Castration resistance in prostate cancer (PCa) constitutes an advanced, aggressive disease with poor prognosis, associated with uncontrolled cell proliferation, resistance to apoptosis, and enhanced invasive potential. The molecular mechanisms involved in the transition of PCa to castration resistance are obscure. Here, we report that the nonselective cationic channel transient receptor potential vanilloid 2 (TRPV2) is a distinctive feature of castration-resistant PCa. TRPV2 transcript levels were higher in patients with metastatic cancer (stage M1) compared with primary solid tumors (stages T2a and T2b). Previous studies of the TRPV2 channel indicated that it is primarily involved in cancer cell migration and not in cell growth. Introducing TRPV2 into androgen-dependent LNCaP cells enhanced cell migration along with expression of invasion markers matrix metalloproteinase (MMP) 9 and cathepsin B. Consistent with the likelihood that TRPV2 may affect cancer cell aggressiveness by influencing basal intracellular calcium levels, small interfering RNA–mediated silencing of TRPV2 reduced the growth and invasive properties of PC3 prostate tumors established in nude mice xenografts, and diminished expression of invasive enzymes MMP2, MMP9, and cathepsin B. Our findings establish a role for TRPV2 in PCa progression to the aggressive castration-resistant stage, prompting evaluation of TRPV2 as a potential prognostic marker and therapeutic target in the setting of advanced PCa. Résumé : Nanosciences and nanotechnologies: health and environment The contribution of the chemist to nanotechnologies is the bottom-up approach, which consists in the assembly of functionalized objects from molecular precursors. The synthesis relies on the supramolecular assembly of elemental bricks, each contributing with a specific functionality. In the case of medical applications, mineral or organic nanoparticles find applications in optical or magnetic imaging as well as in therapy through controlled release and vectorization of drugs. This paper describes both the concept of synthesis and the process methods to elaborate these materials at the industrial scale. Moreover the structured assembly of nano-objects into textured ceramics, hierarchical materials, thin films or active coating favours both surface exchange and reactants diffusion. These systems with open structure find applications in energy conversion, photovoltaic and depollution.

Résumé : Cationic lipophosphoramidates constitute a class of cationic lipids we have previously reported to be efficient for gene transfection. Here, we synthesized and studied a novel lipophosphoramidate derivative characterized by an arsonium headgroup linked, via a phosphoramidate linker, to an unconventional lipidic moiety consisting of two diunsaturated linoleic chains. Physicochemical studies allowed us to comparatively evaluate the specific fluidity and fusogenicity properties of the liposomes formed. Although corresponding lipoplexes exhibited significant but relatively modest in vitro transfection efficiencies, they showed a remarkably efficient and reproducible ability to transfect mouse lung, with in vivo transfection levels higher than those observed with a monounsaturated analogue previously described. Thus, these results demonstrate that this diunsaturated cationic lipophosphoramidate constitutes an efficient and versatile nonviral vector for gene transfection. They also invite further evaluations of the transfection activity, especially in vivo, of gene delivery systems incorporating the lipid reported herein and/or other lipids bearing polyunsaturated chains.

Résumé : There are few data on the prevalence of hypertension in European populations based on the ambulatory determination of blood pressure. The Prognostic Indicator of Cardiovascular and Cerebrovascular Events (PROOF) study provided an opportunity to examine this prevalence in a cohort of 955 patients (387 men and 588 women, all aged 65 years) selected from an electoral list. All patients benefited from measurement of blood pressure during consultation and by 24-hour ambulatory determinations. In this population, 18% had treated hypertension, but 31% had a daytime systolic pressure >135 mm Hg. This proportion was even higher (46%) in patients whose fasting blood glucose was >1.1 g/L or >6.1 mmol/L. In the untreated population, 21% had unrecognized hypertension with daytime systolic pressure >135 mm Hg, with an even higher percentage (35%) in the hyperglycemic patients. This study shows a significant proportion of hypertensive patients, whether treated or not, with high ambulatory blood pressure, especially in those with hyperglycemia who have a higher risk of complications.

Résumé : BACKGROUND: Assisted Reproductive Technologies (ART) are increasingly used in humans; however, their impact is now questioned. At blastocyst stage, the trophectoderm is directly in contact with an artificial medium environment, which can impact placental development. This study was designed to carry out an in-depth analysis of the placental transcriptome after ART in mice. METHODOLOGY/PRINCIPAL FINDINGS: Blastocysts were transferred either (1) after in vivo fertilization and development (control group) or (2) after in vitro fertilization and embryo culture. Placentas were then analyzed at E10.5. Six percent of transcripts were altered at the two-fold threshold in placentas of manipulated embryos, 2/3 of transcripts being down-regulated. Strikingly, the X-chromosome harbors 11% of altered genes, 2/3 being induced. Imprinted genes were modified similarly to the X. Promoter composition analysis indicates that FOXA transcription factors may be involved in the transcriptional deregulations. CONCLUSIONS: For the first time, our study shows that in vitro fertilization associated with embryo culture strongly modify the placental expression profile, long after embryo manipulations, meaning that the stress of artificial environment is memorized after implantation. Expression of X and imprinted genes is also greatly modulated probably to adapt to adverse conditions. Our results highlight the importance of studying human placentas from ART.

Résumé : Vibration sensing by laser radar with a pulsed emission is a promising technique for long range target recognition and identification. However, compared to its continuous-wave counterpart, it is impaired by its greater sensitivity to speckle noise and its lack of robustness to multiple independent vibrations of the target surface. Using poly-pulse waveforms for greater velocity resolution, we developed a new estimator to take into account signal statistics, and time frequency representations that can achieve better performance than classical processing methods. Simulations show a 5dB improvement in Signal-to- Noise Ratio (SNR) when speckle noise is dominant, and 4dB improvement when signal is weak in regard to detection noise.
