Résumé : Apoptosis-Inducing factor (AIF) plays important supportive as well as potentially lethal roles in neurons. Under normal physiological conditions, AIF is a vital redox-active mitochondrial enzyme, whereas in pathological situations, it translocates from mitochondria to the nuclei of injured neurons and mediates apoptotic chromatin condensation and cell death. Here, we reveal the existence of a brain-specific isoform of AIF, AIF2, whose expression increases as neuronal precursor cells differentiate. AIF2 arises from the utilization of the alternative exon 2b, yet uses the same remaining 15 exons as the ubiquitous AIF1 isoform. AIF1 and AIF2 are similarly imported to mitochondria where they anchor to the inner membrane facing the intermembrane space. However, the mitochondrial inner membrane sorting signal (IMSS) encoded in the exon 2b of AIF2 is more hydrophobic than that of AIF1, indicating a stronger membrane anchorage of AIF2 than AIF1. AIF2 is more difficult to be desorbed from mitochondria than AIF1 upon exposure to non-ionic detergents or basic pH. Furthermore, AIF2 dimerizes with AIF1, thereby preventing its release from mitochondria. Conversely, it is conceivable that a neuron-specific AIF isoform, AIF2, may have been "designed" to be retained in mitochondria and to minimize its potential neurotoxic activity.

Résumé : Plio-Pleistocene diatomitic sequences in the Shamb paleo-lake (South Armenia, Lesser Caucasus) offer a rare opportunity to give new insights on the paleo-climate of Western Asia. We present an integrated palynological, 40Ar/39Ar geochronologic and magnetostratigraphic study for the most complete section in the sedimentary deposits of the Shamb paleo-lake. 40Ar/39Ar dating of two volcaniclastic layers provided ages of 1.24 ± 0.03 and 1.16 ± 0.02 Ma (2. Magnetostratigraphic data show that the entire Shamb section is of reversed polarity which, combined with 40Ar/39Ar dating, suggests that the entire section correlates with part of the Matuyama period (1.785-1.070 Ma). Pollen assemblages and macroremains diversity clearly show an alternation of glacial and interglacial phases. Age calibrations and accumulation rate extrapolation allow a direct correlation of climate changes with the global isotopic curve, and show that the Shamb section probably ranges from approximately 1.300 to 1.080 Ma (marine isotopic stages 40 to 31). The vegetation of the Lesser Caucasus developed in a mosaic pattern in a Pleistocene continental, mostly arid climate, comparable to the present-day climate. The observed vegetation changes record a dominant climate response to the obliquity orbital parameter, but the influence of precession could not be established from the Shamb data. Pollen and macroflora both indicate that glacial periods were cold and dry and that interglacials were warm with local humidity. The early Pleistocene climatic model for Western Asia is thus similar to the climatic model for the Mediterranean area.

Résumé : High uptake of HIV voluntary counselling and testing (VCT) services is important for the success of HIV workplace programs in sub-Saharan Africa. From 2001 onwards Heineken, a multinational brewing company, implemented a comprehensive HIV prevention and treatment program for employees and their dependents of its African subsidiaries. Confidential in-house VCT is part of this program. VCT uptake dynamics over time, and factors associated with early uptake were studied. Between September 2001 and December 2007, 9,723 adult beneficiaries were tested for HIV in 14 company sites in 5 African countries. Three-hundred-and-seventy (3.8%) of tested persons were infected with HIV-1. During the first 12 months 1,412 tests were done, compared to 8,311 tests in the subsequent years. The annual average uptake of testing among eligible persons varied between 15% and 32%. The coverage was higher among female compared to male employees, and higher among female compared to male spouses. Distinct peaks in uptake were linked to specific local events. People aged 45 years or above, women, and HIV-1 infected persons were significantly more likely to be tested in the early period. The proportion of HIV-1 infected persons among testees was 8.8% in the first twelve months compared to 3.0% in the subsequent period (p<0.001). HIV-1 infected persons diagnosed in the early period were in a more advanced clinical stage, and had a significantly lower CD4 count than those tested later (median CD4 count 227 vs. 314 cells/μl; p=0.002). In this workplace program HIV-1 infected individuals came earlier for an HIV test than uninfected people, and people with advanced infection came earlier than those with less advanced disease. Employees' spouses are harder to reach than employees and extra efforts should be undertaken to reach them as well. Uptake of HIV testing can be actively influenced by educational or promotional activities.

Résumé : We report on the investigation of strong radiative shocks generated with the high energy, sub-nanosecond iodine laser at PALS. These shock waves are characterized by a developed radiative precursor and their dynamics is analyzed over long time scales (50 ns), approaching a quasi-stationary limit. We present the first preliminary results on the rear side XUV spectroscopy. These studies are relevant to the understanding of the spectroscopic signatures of accretion shocks in Classical T Tauri Stars.

L'invention concerne également un système pour mettre en œuvre ce procédé.

Résumé : PURPOSE: In patients with acute myeloid leukemia (AML), induction chemotherapy is based on standard doses of anthracyclines and cytarabine. High doses of cytarabine have been reported as being too toxic for patients older than age 50 years, but few studies have evaluated intensified doses of anthracyclines. PATIENTS AND METHODS: In this randomized Acute Leukemia French Association 9801 (ALFA-9801) study, high doses of daunorubicin (DNR; 80 mg/m(2)/d x 3 days) or idarubicin (IDA4; 12 mg/m(2)/d x 4 days) were compared with standard doses of idarubicin (IDA3; 12 mg/m(2)/d x 3 days) for remission induction in patients age 50 to 70 years, with an event-free survival (EFS) end point. After two consolidation courses based on intermediate doses of cytarabine, patients in continuous remission were randomly assigned to receive or not receive maintenance therapy with recombinant interleukin-2 (rIL-2; 5 x 10(6) U/m(2) x 5 days each month) for a total duration of 12 months. A total of 468 patients entered the study (median age, 60 years). RESULTS: Overall complete remission rate was 77% with significant differences among the three randomization arms (83%, 78%, and 70% in the IDA3, IDA4, and DNR arms, respectively; P = .04). However, no significant differences were observed in relapse incidence, EFS, or overall survival among the three arms. In the 161 patients randomly assigned for maintenance therapy, no difference in outcome was observed between the rIL-2 and the no further treatment arms. CONCLUSION: Neither intensification of anthracycline doses nor maintenance with rIL-2 showed a significant impact on AML course, at least as scheduled in this trial.

Résumé : What is Stradivari's “secret”? The composition of the mythical varnish that coats Stradivari's violins has raised controversial assumptions for the past two centuries. By using a complementary array of analytical tools, the chemical microstratigraphy of these varnishes has been established. The results provide information on the materials and techniques that were used by the Master, with a detailed characterization of the varnish.
