Résumé : BACKGROUND: Characteristics and factors influencing viral decay under tenofovir (TDF) and adefovir (ADV) need to be determined in HIV-HBV-coinfected patients. METHODS: This open-label study compared the HBV dynamics in 85 HIV-HBV-coinfected patients initiating an antiretroviral regimen, either including TDF or associated with ADV. The first 6-month change in viral load was analysed using mixed linear models. The adjusted hazards ratio, comparing the rates of undetectable HBV DNA between treatments, was calculated using a Cox proportional hazard model. RESULTS: The HBV DNA decay, adjusted for baseline HBV viral load was more pronounced in patients treated with TDF than with ADV at 12 months (66% versus 53%, P=0.0001). Patients in the TDF group presented a steeper slope of decline at 1.1 (95% confidence interval [CI] 0.9-1.3), compared with 0.8 (95% CI 0.6-1.0) in the ADV group (P=0.036). The mean time to HBV DNA undetectability was 19.3 months (95% CI 16.7-22.0) with TDF and 25.9 months (95% CI 21.1-30.7) with ADV. When adjusted for hepatitis B virus e antigen, HBV DNA and alanine aminotransferase levels at baseline, the influence of treatment on time to HBV DNA undetectability remained in favour of TDF versus ADV (hazard ratio=2.79, 95% CI 1.05-7.40, P=0.039) CONCLUSIONS: TDF influenced more strongly the early-phase HBV DNA kinetics than ADV. This is associated with a sustained antiviral activity in the TDF group, in which patients reached the threshold of HBV undetectability at a faster rate and in a larger proportion than those taking ADV.

Résumé : A correlation algorithm is proposed to measure full three-dimensional displacement fields in a three-dimensional domain. The chosen kinematic basis for this measurement is based on continuous finite-element shape functions. It is furthermore proposed to account for the presence of strong discontinuities, similarly to extended finite element schemes, with a suited enrichment of the kinematics with discontinuities supported by a (crack) surface. An optimization of the surface geometry is proposed based on correlation residuals. The procedure is applied to analyze one loading cycle of a fatigue-cracked nodular graphite cast iron sample by using computed tomography pictures. Subvoxel crack openings are revealed and measured.

Résumé : The purpose of this work is to investigate the direct radiative forcing of aerosols over the supersite of Djougou (northern Benin) during the African Monsoon Multidisciplinary Analyses dry season experiment. We focus our simulations on the top of atmosphere, bottom of atmosphere, and atmosphere radiative forcings. During the dry season period, Sun photometer measurements indicate a rather turbid atmosphere with a mean aerosol optical depth for the overall period of 0.78 ± 0.24 (at 440 nm). The aerosol absorption coefficient estimated at the surface ranged between 2.3 and 37.3 Mm−1 (mean value 15.2 ± 10.6 Mm−1 at 520 nm) and the scattering coefficient between 44.5 and 232.3 Mm−1 (mean 145 ± 59 Mm−1 at 520 nm). This leads to a single scattering albedo of between 0.81 and 0.98 (at 520 nm) with a mean (and standard deviation) value of 0.91 ± 0.05, indicating moderately absorbing aerosols. In parallel, micropulse lidar measurements indicate the presence of two distinct aerosol layers, with a first one located between the surface and 1 km and a second one located above 1.5–4.0 km. On the basis of surface and aircraft observations, sunphotometer measurements, lidar profiles, and Moderate Resolution Imagaing Spectroradiometer sensor an estimation of the daily clear sky direct radiative forcing has been estimated for the 17–24 January 2006 period. Simulations indicate that aerosols reduce significantly the solar energy reaching the surface (mean ΔFBOA = −61.5 W/m2) by reflection to space (mean ΔFTOA = −18.4 W/m2) but predominantly by absorption of the solar radiation into the atmosphere (mean ΔFATM = +43.1 W/m2). The mean heating rate at the surface and within the elevated biomass burning layer is considerably enhanced by 1.50 and 1.90 K day−1, respectively.

Résumé : Interactive volume rendering methods such as texture-based slicing techniques and ray casting have been well developed in recent years. The rendering performance is generally restricted by the volume size, the fill-rate, and the texture fetch speed of the graphics hardware. For most 3D data sets, a fraction of the volume is empty, which will reduce the rendering performance without specific optimization. In this paper, we present a simple kd-tree-based space partitioning scheme to efficiently remove the empty spaces from the volume data sets at the preprocessing stage. The splitting rule of the scheme is based on a simple yet effective cost function evaluated through a fast approximation of the bounding volume of the nonempty regions. The scheme culls a large number of empty voxels and encloses the remaining data with a small number of axis-aligned bounding boxes, which are then used for interactive rendering. The number of boxes is controlled by halting criteria. In addition to its simplicity, our scheme requires little preprocessing time and improves the rendering performance significantly.

Résumé : Among vertebrate model species, the zebrafish embryo combines at an unprecedented level optical accessibility with easy genetic manipulation. As such, it is gaining recognition as a powerful model to study innate immunity. In this chapter, we provide a protocol for the generation of zebrafish embryos deficient in a protein of interest for innate immune signaling using antisense morpholino oligonucleotides, the systemic or local infection of these embryos with bacteria, and the assessment of various aspects of the following immune response with emphasis on microscopic observation. This example can be easily adapted to study the role of other genes, either knocked down or overexpressed, and in response to any other challenge, from purified microbial compounds to pathogenic viruses. This protocol is aimed at people not necessarily familiar with zebrafish biology and handling.
