Résumé : Glial Ca(2+) excitability plays a key role in reciprocal neuron-glia communication. In the retina, neuron-glia signalling is expected to be maximal in the dark, but the glial Ca(2+) signal characteristics under such conditions have not been evaluated. To address this question, we used bioluminescence imaging to monitor spontaneous Ca(2+) changes under dark conditions selectively in M?r cells, the principal retinal glial cells. By combining this imaging approach with network analysis, we demonstrate that activity in M?r cells is organized in networks of coactive cells, involving 2-16 cells located distantly and/or in clusters. We also report that spontaneous activity of small networks (2-6 M?r cells) repeat over time, sometimes in the same sequential order, revealing specific temporal dynamics. In addition, we show that networks of coactive glial cells are inhibited by TTX, indicating that ganglion and/or amacrine neuronal cells probably regulate M?r cell network properties. These results represent the first demonstration that spontaneous activity in adult M?r cells is patterned into correlated networks that display repeated sequences of coactivations over time. Furthermore, our bioluminescence technique provides a novel tool to study the dynamic characteristics of glial Ca(2+) events in the retina under dark conditions, which should greatly facilitate future investigations of retinal dark-adaptive processes.

Résumé : High-dose chemotherapy followed by autologous hematopoietic stem-cell transplantation has been reported to provide higher response rates and better overall survival than standard chemotherapy in immunoglobulin-light-chain (AL) amyloidosis, but these two strategies have not been compared in a randomized study. METHODS: We conducted a randomized trial comparing high-dose intravenous melphalan followed by autologous hematopoietic stem-cell rescue with standard-dose melphalan plus high-dose dexamethasone in patients with AL amyloidosis. Patients (age range, 18 to 70 years) with newly diagnosed AL amyloidosis were randomly assigned to receive intravenous high-dose melphalan plus autologous stem cells or oral melphalan plus oral high-dose dexamethasone. RESULTS: Fifty patients were enrolled in each group. The results were analyzed on an intention-to-treat basis, with overall survival as the primary end point. After a median follow-up of 3 years, the estimated median overall survival was 22.2 months in the group assigned to receive high-dose melphalan and 56.9 months in the group assigned to receive melphalan plus high-dose dexamethasone (P=0.04). Among patients with high-risk disease, overall survival was similar in the two groups. Among patients with low-risk disease, there was a nonsignificant difference between the two groups in overall survival at 3 years (58% in the group assigned to receive high-dose melphalan vs. 80% in the group assigned to receive melphalan plus high-dose dexamethasone; P=0.13). CONCLUSIONS: The outcome of treatment of AL amyloidosis with high-dose melphalan plus autologous stem-cell rescue was not superior to the outcome with standard-dose melphalan plus dexamethasone. Résumé : This paper describes the design of a drive consisting of a DC motor, a speed reducer, a lead screw transformation system, a power converter and its associated DC source. The objective is to reduce the mass of the system. Indeed, the volume and weight optimisation of an electrical drive is an important issue for embedded applications. Here, we present an analytical model of the system in a specific application and afterwards an optimisation of the motor and speed reducer main dimensions and the battery voltage in order to reduce the weight.

Résumé : This paper concerns the integration of a complex vision algorithm on a dedicated architecture. Perception is required on a vehicle (robot or car), especially in order to detect obstacles along a trajectory for a robot or on the road for a car. This function must satisfy very hard real-time constraints so that an avoidance strategy could be executed by the onboard system on a robot or by a driver on a car: it is required to detect obstacles at 100Hz. Several sensors have been used to acquire perceptual data from which obstacles are detected: we propose to use a stereovision sensor. This algorithm is so complex that it must be integrated on a dedicated architecture to provide a high resolution 3D reconstruction of the environment at 100Hz. At first, this paper describes a correlation-based stereovision algorithm, and a generic architecture including several FPGA-based boards, designed to execute visual functions at very high frequency. Then we propose a strategy to implement our stereovision algorithm on this computing unit, with four FPGA-based boards.

Résumé : In this paper we present a new method of data hiding in 3D objects. This method is based on a 3D model represented by a cloud of vertices and a list of edges corresponding to the triangular mesh of the surface. The main idea of this method is to find and to synchronize particular areas that can be used to embed the message. The data hiding relies on the modification of the topology of edges in chosen areas. These modifications, as a consequence, have to change the structure of triangles constructed in these areas. This method has the advantage of not changing the position of the vertices of the 3D model. This invariant on the vertex positions allows us to carry out an embedding that is robust to the affine transformations of type rotation, translation or zoom.

Résumé : We study regression estimation when the explanatory variable is functional. Nonparametric estimates of the regression operator have been recently introduced. They depend on a smoothing factor which controls its behavior, and the aim of our work is to construct some data-driven criterion for choosing this smoothing parameter. The criterion can be formulated in terms of a functional version of cross-validation ideas. Under mild assumptions on the unknown regression operator, it is seen that this rule is asymptotically optimal. As by-products of this result, we state some asymptotic equivalences for several measures of accuracy for nonparametric estimate of the regression operator. We also present general inequalities for bounding moments of random sums involving functional variables. Finally, a short simulation study is carried out to illustrate the behavior of our method for finite samples.
