Résumé : BACKGROUND: Increasing evidence supports the usefulness of brain magnetic resonance imaging (MRI) for the diagnosis of human prion diseases. From the neuroradiological point of view, fatal familial insomnia is probably the most challenging to diagnose because brain lesions are mostly confined to the thalamus. OBJECTIVE: To determine whether multisequence MRI of the brain can show thalamic alterations and establish pathoradiologic correlations in a patient with familial fatal insomnia. DESIGN: Radioclinical prospective study. We describe a patient with fatal familial insomnia and normal MRI images. Because the MRI study was performed only 4 days before the patient's death, we were able to compare radiological data with the lesions observed at the neuropathologic level. Patient A 55-year-old man with familial fatal insomnia. Main Outcome Measure Magnetic resonance spectroscopy combined with the measurement of apparent diffusion coefficient of water in different brain areas. RESULTS: The neuroradiological study showed, in the thalamus but not in the other brain regions studied, an increase of apparent diffusion coefficient of water and a metabolic pattern indicating gliosis. These alterations closely correlated with neuropathologic data showing an almost pure gliosis that was restricted to the thalami. CONCLUSION: Considering fatal familial insomnia as a model of thalamic-restricted gliosis, this case demonstrates that multisequences of magnetic resonance can detect prion-induced gliosis in vivo, as confirmed by a neuropathologic examination performed only a few days after radiological examination.

Résumé : Ce travail présente les caractéristiques technotypologiques de l’industrie lithique de deux sites de plein air, associés à un banc de silex oligocène. Deux phases d’occupation sont présentes sur place. La première présente les caractéristiques de l’Épipaléolithique et se caractérise par la production de supports laminaires obtenus au percuteur de pierre tendre, à partir de nucléus mis en forme. La réalisation de préformes de nucléus pourraient également se rattacher à cette phase culturelle. La seconde période de fréquentation du site date du Mésolithique. Le débitage est orienté vers la production de supports lamellaires détachés au percuteur de pierre tendre, à partir de petits nucléus qui n’ont subi aucune mise en forme. Parallèlement à l’approche technologique de l’industrie, ce sont les comportements humains liés aux différentes modalités d’exploitation du banc de silex qui sont appréhendés.

Résumé : Previous studies of the HIV-1 disease have shown that HLA and Chemokine receptor genetic variants influence disease progression and early viral load. We performed a Genome Wide Association study in a cohort of 605 HIV-1-infected seroconverters for detection of novel genetic factors that influence plasma HIV-RNA and cellular HIV-DNA levels. Most of the SNPs strongly associated with HIV-RNA levels were localised in the 6p21 major histocompatibility complex (MHC) region and were in the vicinity of class I and III genes. Moreover, protective alleles for four disease-associated SNPs in the MHC locus (rs2395029, rs13199524, rs12198173 and rs3093662) were strikingly over-represented among forty-five Long Term HIV controllers. Furthermore, we show that the HIV-DNA levels (reflecting the HIV reservoir) are associated with the same four SNPs, but also with two additional SNPs on chromosome 17 (rs6503919; intergenic region flanked by the DDX40 and YPEL2 genes) and chromosome 8 (rs2575735; within the Syndecan 2 gene). Our data provide evidence that the MHC controls both HIV replication and HIV reservoir. They also indicate that two additional genomic loci may influence the HIV reservoir.

Résumé : Background. The evolution of the variant Creutzfeldt-Jakob disease (vCJD) epidemic is hazardous to predict due to uncertainty in ascertaining the prevalence of infection and because the disease might remain asymptomatic or produce an alternate, sporadic-like phenotype. Methodology/Principal Findings. Transgenic mice were produced that overexpress human prion protein with methionine at codon 129, the only allele found so far in vCJD-affected patients. These mice were infected with prions derived from variant and sporadic CJD (sCJD) cases by intracerebral or intraperitoneal route, and transmission efficiency and strain phenotype were analyzed in brain and spleen. We showed that i) the main features of vCJD infection in humans, including a prominent involvement of the lymphoid tissues compared to that in sCJD infection were faithfully reproduced in such mice; ii) transmission of vCJD agent by intracerebral route could lead to the propagation of either vCJD or sCJD-like prion in the brain, whereas vCJD prion was invariably propagated in the spleen, iii) after peripheral exposure, inefficient neuroinvasion was observed, resulting in an asymptomatic infection with life-long persistence of vCJD prion in the spleen at stable and elevated levels. Conclusion/Significance. Our findings emphasize the possibility that human-to-human transmission of vCJD might produce alternative neuropathogical phenotypes and that lymphoid tissue examination of CJD cases classified as sporadic might reveal an infection by vCJD-type prions. They also provide evidence for the strong propensity of this agent to establish long-lasting, subclinical vCJD infection of lymphoreticular tissues, thus amplifying the risk for iatrogenic transmission.

Résumé : Various iodo-acridone and acridine carboxamides have been prepared and evaluated as agents for targeted radionuclide and/or chemotherapy for melanoma, due to their structural similarity to benzamides which are known to possess specific affinity for melanin. Three of these carboxamides selected for their in vitro cytotoxic properties were radioiodinated with [125I]NaI at high specific activity. Biodistribution studies carried out in B16F0 murine melanoma tumour-bearing mice highlighted that acridone 8f and acridine 9d, presented high, long-lasting tumour concentrations together with an in vivo kinetic profile favourable to application in targeted radionuclide therapy.

Résumé : The impact of stratospheric ozone on the tropospheric general circulation of theSouthern Hemisphere (SH) is examined with a set of chemistry‐climate models participatingin the Stratospheric Processes and their Role in Climate (SPARC)/Chemistry‐ClimateModel Validation project phase 2 (CCMVal‐2). Model integrations of both the past andfuture climates reveal the crucial role of stratospheric ozone in driving SH circulationchange: stronger ozone depletion in late spring generally leads to greater polewarddisplacement and intensification of the tropospheric midlatitude jet, and greater expansion ofthe SH Hadley cell in the summer. These circulation changes are systematic as polewarddisplacement of the jet is typically accompanied by intensification of the jet and expansion ofthe Hadley cell. Overall results are compared with coupled models participating in theIntergovernmental Panel on Climate Change Fourth Assessment Report (IPCC AR4), andpossible mechanisms are discussed. While the tropospheric circulation response appearsquasi‐linearly related to stratospheric ozone changes, the quantitative response to a givenforcing varies considerably from one model to another. This scatter partly results fromdifferences in model climatology. It is shown that poleward intensification of the westerly jetis generally stronger in models whose climatological jet is biased toward lower latitudes.This result is discussed in the context of quasi‐geostrophic zonal mean dynamics.
