Résumé : OBJECTIVE: CD4(+)CD25(+) regulatory T lymphocytes (Treg) have been initially shown to prevent organ-specific autoimmunity. It is now accepted that Treg homeostasis depends in part on the peripheral conversion of na? CD4(+)CD25(-) T cells. This conversion implicates acquisition of the Treg-specific markers, forkhead winged helix protein 3 (Foxp3), after CD28 costimulation. Because natural killer cells (NK) are critical for efficient cytotoxic T-cell priming and TH1 polarization, we investigated their role in Foxp3 induction in CD4(+) T lymphocytes. MATERIALS AND METHODS: Human CD4(+)CD25(-) T lymphocytes were activated in vitro by CD28 costimulation in the presence of interleukin-2-activated NK. Three days after initial activation, Foxp3 protein and RNA expression were determined by flow cytometry and reverse transcription polymerase chain reaction. In vivo influence of activated NK on Foxp3 expression was studied in an adoptive transfer model of CD45.2(+) CD4(+)CD25(-) lymphocytes into CD45.1(+) mice. RESULTS: Interleukin-2-activated NK decreased Treg conversion of adoptively transferred murine CD4(+)CD25(-) T cells in vivo. Likewise, human-activated NK, but not resting NK, decreased CD28-driven Foxp3 expression in CD4(+)CD25(-) T lymphocytes, while at the same time increasing proliferation and interferon-gamma (IFN-gamma) production. Neutralization of IFN-gamma partially restored Treg conversion and prevented TH1 polarization after CD28 costimulation. CONCLUSION: The current study suggests that activated NK interfere with CD28-mediated Foxp3 expression in CD4(+)CD25(-) T lymphocytes. Our experiments further underline a molecular interaction between IFN-gamma and Foxp3 downstream of CD28 signaling. Together, these results demonstrate that activated NK play a critical role at the initiation step of immune responses by modulating peripheral Treg differentiation.

Résumé : Si les régions contribuent aux investissements affectés aux réseaux routiers et à l'extension du réseau TGV, elles sont aussi depuis le début des années 2000 autorités organisatrices des transports régionaux de voyageurs (TER). Sept équipes de chercheurs ont mené en commun, sous l'égide du PREDIT (GO 11), une enquête comparative et pluridisciplinaire en 2006 dans les régions Nord - Pas de Calais, Rhône-Alpes, Alsace, Midi-Pyrénées, PACA et Centre. La recherche a débouché sur un certain nombre de résultats sur les mécanismes d'institutionnalisation régionale, l'émergence de nouveaux corps de doctrine, le processus de différenciation territoriale, l'évolution des réformes de décentralisation et la montée en puissance des pouvoirs métropolitains?

Résumé : We report the first experimental observation of reversals of a dynamo field generated in a laboratory experiment based on a turbulent flow of liquid sodium. The magnetic field randomly switches between two symmetric solutions B and -B. We observe a hierarchy of time scales similar to the Earth's magnetic field: the duration of the steady phases is widely distributed, but is always much longer than the time needed to switch polarity. In addition to reversals we report excursions. Both coincide with minima of the mechanical power driving the flow. Small changes in the flow driving parameters also reveal a large variety of dynamo regimes.

Résumé : Background : Epidemiologic data suggest that infection with herpes simplex virus type 2 (HSV-2) is associated with increased genital shedding of human immunodeficiency virus type 1 (HIV-1) RNA and HIV-1 transmissibility. Methods : We conducted a randomized, double-blind, placebo-controlled trial of HSV suppressive therapy with valacyclovir (at a dose of 500 mg twice daily) in Burkina Faso among women who were seropositive for HIV-1 and HSV-2; all were ineligible for highly active antiretroviral therapy. The patients were followed for 24 weeks (12 weeks before and 12 weeks after randomization). Regression models were used to assess the effect of valacyclovir on the presence and quantity of genital and plasma HIV-1 RNA and genital HSV-2 DNA during treatment, adjusting for baseline values, and to evaluate the effect over time. Results : A total of 140 women were randomly assigned to treatment groups; 136 were included in the analyses. At enrollment, the median CD4 cell count was 446 cells per cubic millimeter, and the mean plasma viral load was 4.44 log10 copies per milliliter. With the use of summary-measures analysis, valacyclovir therapy was found to be associated with a significant decrease in the frequency of genital HIV-1 RNA (odds ratio, 0.41; 95% confidence interval [CI], 0.21 to 0.80) and in the mean quantity of the virus (log10 copies per milliliter, −0.29; 95% CI, −0.44 to −0.15). However, there was no significant decrease in detection of HIV (risk ratio, 0.93; 95% CI, 0.81 to 1.07). HSV suppressive therapy also reduced the mean plasma HIV-1 RNA level by 0.53 log10 copy per milliliter (95% CI, −0.72 to −0.35). Repeated-measures analysis showed that these effects became significantly stronger during the 3 months of follow-up. Conclusions : HSV suppressive therapy significantly reduces genital and plasma HIV-1 RNA levels in dually infected women. This finding may have important implications for HIV control. Résumé : The alternate deposition of exponentially and linearly growing polyelectrolyte multilayers leads to the formation of multicompartment films. In this study, a new system consisting in nanometer-sized multilayer barriers deposited on or between multilayer compartments was designed to respond to mechanical stimuli and to act as nanovalves. The diffusion of polyelectrolytes through the barrier from one compartment to another can be switched on/off by tuning the mechanical stretching and thereby opening or closing nanopores in the barrier. This work represents a first step toward the design of chemically or biologically active films responding to mechanical stresses.

Résumé : The goals of the present contribution are twofold. First, we propose the use of a non-Gaussian long-range dependent process to model Internet traffic aggregated time series. We give the definitions and intuition behind the use of this model. We detail numerical procedures that can be used to synthesize artificial traffic exactly following the model prescription. We also propose original and practically effective procedures to estimate the corresponding parameters from empirical data. We show that this empirical model relevantly describes a large variety of Internet traffic, including both regular traffic obtained from public reference repositories and traffic containing legitimate (flash crowd) or illegitimate (DDoS attack) anomalies. We observe that the proposed model accurately fits the data for a wide range of aggregation levels. The model provides us with a meaningful multiresolution (i.e., aggregation level dependent) statistics to characterize the traffic: the evolution of the estimated parameters with respect to the aggregation level. It opens the track to the second goal of the paper: anomaly detection. We propose the use of a quadratic distance computed on these statistics to detect the occurrences of DDoS attack and study the statistical performance of these detection procedures. Traffic with anomalies was produced and collected by us so as to create a controlled and reproducible database, allowing for a relevant assessment of the statistical performance of the proposed (modeling and detection) procedures.

Résumé : La matière organique des sols et leur gestion sont des facteurs importants de production. Selon le mode d'exploitation forestière plus ou moins de rémanents (feuilles, branches, écorces, bois) retournent sur le sol. Un essai sur l’impact de la gestion des résidus d'exploitation (rémanents et litière préexistante) sur la croissance d'une replantation d’Eucalyptus a été mis en place en 1998 dans la région de Pointe-Noire (Congo) où les sols sableux sont très pauvres en matière organique et en éléments minéraux. Cet essai compare 6 modes de gestion des résidus d'exploitation sur l’évolution de la croissance des Eucalyptus et sur la fertilité des sols. Ces 6 traitements permettent d'avoir une gradation des quantités de résidus de 0 à 49.4 tMS.ha-1 dont un traitement de référence (la pratique usuelle du gestionnaire des plantations) et un traitement où les résidus ont été brûlés avant plantation. Le suivi de cet essai pendant 7 ans révèle que la croissance des Eucalyptus est immédiatement et fortement influencée par la gestion des résidus d'exploitation. La production en volume à 7 ans est de 84 m3.ha-1 pour le traitement où tous les résidus ont été enlevés, de 146 m3.ha-1 pour le témoin, de 161 m 3.ha-1 pour le traitement où la quantité de résidus a été doublée et de 127 m3.ha-1 pour le traitement où les résidus ont été brûlés. Les résultats d'analyses foliaires, de chutes de litière et de minéralisation montrent que, dans ces sols très pauvres, les Eucalyptus se nourrissent en grande partie à partir du recyclage rapide des matières organiques et grâce à un mat racinaire superficiel. L’évolution des sols indique des modifications notables de la saturation, malgré la fertilité très faible du sol. Ce site appartient à un réseau de 17 sites expérimentaux répartis de la Chine au Brésil en zone intertropicale, où les mêmes traitements ont été appliqués. Ce réseau est coordonné par le Centre International des Recherches Forestières (CIFOR). L’utilisation de la base de données du réseau pour modéliser de l’effet du traitement des rémanents montre que la baisse initiale de production peut ou non disparaître avec le temps, en fonction de la fertilité initiale des sols et à leur capacité de restauration. Ces 24 résultats ont une portée très générale pour discuter de l’effet initial et à long terme du traitement des rémanents en forêt, quel que soit le type de climat.
