Résumé : Tourist sites in natural areas constitute a remarkable subjet of study for better understanding the way in which the local area or territory participates in the production of a localised cultural system (LCS). Indeed, while the economic function of such sites consists in producing weath for the entrepreneurs proposing multiple services to the tourists present ont he sites, it seems simplistic to consider a touriste destination as being no more than a simple aggregation of actors. The existence of ties with the territory that can be qualified locally points to the presence of a cultural process that plays an important role in the creation of resources that are essential to the efficient operation of such a system. Drawing on several examples of sports and tourist sites in the French Alps, we show how this localised cultural system in built up and how it takes shape. The study of the forms of development st up by the different service providers and of the sports culture of the tourists provides valuable insights into the make-up and practical organisation of these cultural resources in an outdoor recreational site.

Résumé : Ami is an autolytic amidase from Listeria monocytogenes that is targeted to the bacterial surface via its C-terminal cell wall anchoring (CWA) domain. We recently showed that the CWA domain from Ami of L. monocytogenes EGD (serovar 1/2a) (Ami 1/2a) mediated bacterial binding to mammalian cells. Here we studied the sequence and binding properties of Ami from CHUT 82337 (serovar 4b) (Ami 4b). The Ami 4b polypeptide is predicted to be 770 amino acids long (compared with the 917 amino acids of Ami 1/2a from EGD). Ami 1/2a and Ami 4b are almost identical in the N-terminal enzymatic domain (~98% amino acid identity), but the sequence is poorly conserved in the C-terminal CWA domain, with only ~54% amino acid identity and eight GW modules in Ami 1/2a compared with six GW modules in Ami 4b. The purified Ami 4b CWA domain efficiently bound serovar 4b bacterial cells and only poorly bound serovar 1/2a bacterial cells. The Ami 4b CWA domain was also significantly less able to bind Hep-G2 human hepatocytic cells than the Ami 1/2a CWA domain. We sequenced the ami regions encoding CWA domains of reference strains belonging to the 12 L. monocytogenes serovars. The phylogenic tree constructed from the sequences yielded a binary division into group I (serovars 1/2a, 1/2b, 1/2c, 3a, 3b, 3c, and 7) and group II (serovars 4a, 4b, 4c, 4d, and 4e). This is the first direct evidence of divergence between serovars 1/2a and 4b in a gene involved in the adhesion of L. monocytogenes to mammalian cells, as well as the first demonstration of allelic polymorphism correlated with the somatic antigen in this species.

Résumé : This document presents our very first results on a part of the program: "A study of the Be phenomenon in young open Galactic and Magellanic clusters". It concerns the B stars population in the LMC NGC2004 cluster and its surrounding field observed with the VLT/FLAMES instrumentation with the GIRAFFE spectrograph, at low resolution, in MEDUSA mode. First observational results which concern the ratio $\frac{Be}{B+Be}$ , the nebulosities in the field and the discovery of spectrocopic binaries are presented.

Résumé : Whereas Glc is stored in small-sized hydrosoluble glycogen particles in archaea, eubacteria, fungi, and animal cells, photosynthetic eukaryotes have resorted to building starch, which is composed of several distinct polysaccharide fractions packed into a highly organized semicrystalline granule. In plants, both the initiation of polysaccharide synthesis and the nucleation mechanism leading to formation of new starch granules are currently not understood. Ostreococcus tauri, a unicellular green alga of the Prasinophyceae family, defines the tiniest eukaryote with one of the smallest genomes. We show that it accumulates a single starch granule at the chloroplast center by using the same pathway as higher plants. At the time of plastid division, we observe elongation of the starch and division into two daughter structures that are partitioned in each newly formed chloroplast. These observations suggest that in this system the information required to initiate crystalline polysaccharide growth of a new granule is contained within the preexisting polysaccharide structure and the design of the plastid division machinery.

Résumé : Un programme de cherche national entre l`ONF, l`ONCFS et le Cemagref a été initié pour connaître les effets de la sylviculture sur les populations de chevreuils. Les outils de suivis mis en place et les premiers résultats de ce travail sont présentés au cas particulier d`une chênaie (forêt de Trois fontaines - 52) qui se caractérise par une abondante population de chevreuils suivie depuis presque 30 ans.
