Résumé : Crucial events in myogenesis rely on the highly regulated spatiotemporal distribution of cell surface heparan sulfate proteoglycans to which are associated growth factors, thus creating a specific microenvironment around muscle cells. Most growth factors involved in control of myoblast growth and differentiation are stored in the extracellular matrix through interaction with specific sequences of glycosaminoglycan oligosaccharides, mainly heparan sulfate (HS). Different HS subspecies revealed by specific antibodies, have been shown to provide spatiotemporal regulation during muscle development. We have previously shown that glycosaminoglycan (GAG) mimetics called RGTA (ReGeneraTing Agent), stimulate muscle precursor cell growth and differentiation. These data suggest an important role of GAGs during myogenesis; however, little is yet known about the different species of GAGs synthesized during myogenesis and their metabolic regulation. We therefore quantified GAGs during myogenesis of C2.7 cells and show that the composition of GAG species was modified during myogenic differentiation. In particular, HS levels were increased during this process. In addition, the GAG mimetic RGTA, which stimulated both growth and differentiation of C2.7 cells, increased the total amount of GAG produced by these cells without significantly altering their rate of sulfation. RGTA treatment further enhanced HS levels and changed its sub-species composition. Although mRNA levels of the enzymes involved in HS biosynthesis were almost unchanged during myogenic differentiation, heparanase mRNA levels decreased. RGTA did not markedly alter these levels. Here we show that the effects of RGTA on myoblast growth and differentiation are in part mediated through an alteration of GAG species and provide an important insight into the role of these molecules in normal or pathologic myogenic processes.

Résumé : We report the first case of a human neuromuscular transmission dysfunction due to mutations in the gene encoding the muscle-specific receptor tyrosine kinase (MuSK). Gene analysis identified two heteroallelic mutations, a frameshift mutation (c.220insC) and a missense mutation (V790M). The muscle biopsy showed dramatic pre- and postsynaptic structural abnormalities of the neuromuscular junction and severe decrease in acetylcholine receptor (AChR) epsilon-subunit and MuSK expression. In vitro and in vivo expression experiments were performed using mutant MuSK reproducing the human mutations. The frameshift mutation led to the absence of MuSK expression. The missense mutation did not affect MuSK catalytic kinase activity but diminished expression and stability of MuSK leading to decreased agrin-dependent AChR aggregation, a critical step in the formation of the neuromuscular junction. In electroporated mouse muscle, overexpression of the missense mutation induced, within a week, a phenotype similar to the patient muscle biopsy: a severe decrease in synaptic AChR and an aberrant axonal outgrowth. These results strongly suggest that the missense mutation, in the presence of a null mutation on the other allele, is responsible for the dramatic synaptic changes observed in the patient.

Résumé : The main goal of this paper is to define, study and analyse a remote control architecture for a set of non- holonomic robotic vehicles. This project (ACCORD) gathers three laboratories and the French Army Re- search Office. Each of these laboratories deals with a part of this multidisciplinary project which in- cludes coordinated control, control architecture, con- trol with time delay and monitoring of the wireless network. In this paper, we propose to present the whole goal of this project including the basis experi- mental setup developed to validate our control algo- rithm. Secondly, we focuse on a new decentralized control strategy that uses the Leader-Follower prin- ciple. The originality of this paper stems from the use of the signal level of the wireless connection as a control vector. Indeed, each vehicle is fitted with two wireless devices. One of them is equipped of a sector antenna fitted on DC-motor to track the direction of better receiving level. Thus, it allows us to know the relative angular position of the Follower pointing out the Leader. By using the wireless technology as sensor instead of vision for instance, allows a longer distance of coordinated control loop between each vehicle (approx. 100m) even if the GPS information is not available.

Résumé : Within the context of sustainable management of forest ecosystems, a network was started by the Center for International Forestry Research in 1995 to evaluate the impacts of slash management practices on the productivity of eucalypt, acacia and pine plantations. This network has now reached one full rotation for the ten eucalypt sites in Australia, Brazil, China, Congo, India and South Africa. This paper reports an overall synthesis of tree and stand growth. Using a growth and yield modelling approach, we investigated the effects of slash management on site index defined as the asymptotic dominant height, basal area growth and stand structure. There were significant effects of slash and litter management treatments on two processes that drive tree and stand growth: site index and ability of trees to capture the site potential. These impacts were strongly site dependent. As expected, the soil organic carbon concentration could not be used alone to predict the intensity of stand response. Conversely, a ratio between nitrogen content in slash and litter prior to planting and the nitrogen concentration within the 0-10 cm soil layer, was a good predictor of the differences in stand productivity across treatments.

Résumé : The amino-zinc-ene-enolate cyclization reaction is a straightforward route to the synthesis of 3-substituted prolines. Herein we report the application of this reaction to the syntheses of proline chimeras of lysine, glutamic acid, glutamine, arginine, and serine. All these compounds were obtained in enantiomerically pure form and suitably protected for peptide synthesis.
