Résumé : AMBER is a focal instrument for the Very Large Telescope Interferometer working in the near infrared from 1.1 to 2.4 micrometers . It has been designed having in mind the General User of interferometric observations and the full range of his possible astrophysical programs. However the three programs used to define the key specifications have been the study of Young Stellar Objects, the study of Active Galactic Nuclei dust tori and broad line regions and the measure of masses and spectra of hot Extra Solar Planets. AMBER combines up to three beams produced by the VLTI 8 m Unit Telescopes equipped with Adaptive Optics and/or by the 1.8 m Auxiliary Telescopes. The fringes are dispersed with resolutions ranging from 35 to 10000. It is optimized for high accuracy single mode measurements of the absolute visibility, of the variation of the visibility and phase with wavelength (differential interferometry) and of phase closure relations with three telescopes. The instrument and its software are designed to allow a highly automated user friendly operation and an easy maintenance.

Résumé : Mesogenic compounds containing four rings in the core usually have very high melting points. However, when two identical lateral benzyloxy groups are introduced on the same side of one of the central rings, the melting point is lowered dramatically and a large nematic range is retained. This range is affected by the bulkiness of the para-substituents in the lateral rings. Methyl groups can be introduced in the ortho- or meta-positions with a consequent decrease in the melting temperature without much affecting the nematic range. These compounds exhibit a rich solid polymorphism which is certainly related to the effect of the conformations of the lateral substituent on the molecular arrangment in the solid phase. Some preliminary NMR experiments on the nematic phase indicate that the molecular long axis coincides with the core axis, whereas the para-axis of the lateral fragment makes an angle close to the magic angle with respect to the molecular long axis.

Résumé : The synthesis of organic acids was studied during fruit development of two peach (Prunus persica L, Batsch) cultivars, Fantasia and Jalousia, having fruits with high and low organic acid content, respectively. The malate content was higher in cv. Fantasia than in cv, Jalousia at the end of the first rapid growth stage (50 days after bloom [DAB]). Malate and citrate contents, were higher in Fantasia than in Jalousia during the second rapid growth stage (from 100 DAB to maturity). The expression of phosphoenolpyruvate carboxylase (PEPC, EC 4.1.1.31), which is involved in organic acid synthesis, was studied during peach fruit development. PEPC mRNA levels, and protein levels on a total soluble protein basis, peaked at 23 and 108 DAB in Fantasia. In Jalousia, they were very low at 23 DAB and reached levels similar to Fantasia at 108 DAB, For both cultivars, in vitro PEPC activity expressed on a dry weight basis was maximal at 24 DAB, decreased from 24 to 60 DAB, and then remained constant. The activity of peach fruit PEPC appeared extremely sensitive to malate (I-0.5 of 100 mu M for Fantasia and 65 mu M for Jalousia at pH 7.3) and low pH, PEPC may participate in the control of organic acid accumulation during fruit development in the normal-acid fruit of Fantasia, However, mechanisms other than organic acid synthesis might account for the differences in acidity between normal-acid and non-acid peach fruit.

Résumé : Background: TT virus (TTV) is a recently discovered virus, which is not related to any other known virus infecting humans. Objectives: To investigate: (i) the world-wide distribution of the three major TTV genotypes; and (ii) the possible routes of viral transmission. Study design: (i) The phylogenetic distribution of 494 TTV isolates originating from 31 countries was analysed, using partial ORF1 sequences. (ii) Faeces samples (n=22) and saliva samples (n=72) from French individuals were tested for the presence of TTV DNA. (iii) Viral titres in paired serum and saliva samples were compared. Results: (i) Genotypes 1, 2 and 3 were distributed world-wide, with a high proportion of type 1 in Asia (71%) and no type 3 identified in Africa to date. In the USA, 77% of isolates were grouped in four clusters only (genetic distances <10%). This was also the case of 76% of French isolates, 76% of Japanese isolates, and 89% of Hong Kong isolates. (ii) TTV DNA was detected in 18% of faeces samples and 68% of saliva samples tested. (iii) Viral titre in saliva samples was 100-1000 times higher than that of the corresponding serum. Conclusions: (i) The observed epidemiological distribution of TTV isolates is compatible with an ancient dissemination of viral ancestors belonging to the different genotypes and a slow genetic evolution in sedentary populations. (ii) Besides the possible transmission of TTV by the parental and oral-faecal routes, the high titre of TTV DNA observed in saliva raises the hypothesis of the viral transmission by saliva droplets. This route of transmission could explain the high degree of exposure to viral infection observed in the general population.
