Résumé : Introduction: The environmental impact of endoscopy, including small-bowel capsule endoscopy (SBCE), has gained attention due to its contribution to the global carbon footprint. This study aimed to evaluate the greenhouse gas (GHG) emissions (kgCO2e) of SBCE, including devices life cycle and capsule journey. Methods: SBCE devices (3 brands) were evaluated using life cycle assessment methodology (ISO 14040), including patient travelling, bowel preparation, capsule examination and video recording. A survey was conducted on 120 patients undergoing a SBCE to gather data on their transportation, activities during the procedure, and awareness of pollution generated and on 87 physicians reading capsules. Results: For the 3 different capsules, the weight was 4 g (3.9-5.2% of total), while 43 to 119 g were attributed for packaging (9-97%) including 5 g of deactivation magnets (4-6%) and 11 to 50 g for instruction forms (40%). A full SBCE generated between 19 and 20 kgCO2e, including 0.04 kgCO2e (0.2%) for the capsule itself and 18 kgCO2e related to patient travelling (94.7%). Capsule retrieval would add 0.98 kgCO2e using dedicated devices. Capsule deconstruction revealed components (e.g. Neodymium) that are prohibited from environmental disposal. 76% of patients were not aware of the illegal nature of flushing capsules, and 63% would be willing to retrieve it. Data storage and physician impact were negligible. Conclusion: The GHG emission of SBCE is mainly determined by patient travelling. The capsule device itself has a comparably low carbon footprint. Considering capsule components disposal is illegal, retrieval of the capsule seems crucial but increasing device-related emissions.

Résumé : IMPORTANCE
There is still considerable controversy in the literature regarding the capacity of intramuscular messenger RNA (mRNA) vaccination to induce a mucosal immune response. OBJECTIVE To compare serum and salivary IgG and IgA levels among mRNA-vaccinated individuals with or without previous SARS-CoV-2 infection. DESIGN, SETTING, AND PARTICIPANTS In this cohort study, SARS-CoV-2-naive participants and those with previous infection were consecutively included in the CoviCompare P and CoviCompare M mRNA vaccination trials and followed up to day 180 after vaccination with either the BNT162b2 (Pfizer-BioNTech) vaccine or the mRNA-1273 (Moderna) vaccine at the beginning of the COVID-19

Résumé : Botryosphaeria dieback is a grapevine trunk disease caused by fungi of the Botryosphaeriaceae family, which attacks more specifically the woody tissues. The infection leads to different symptoms including a severe form with a leaf drop as well as premature plant death. Botryosphaeria dieback causes major economic losses, since no effective treatment is yet available. A better understanding is necessary to find solutions to fight this disease. In this study, our objective was to characterize the “leaf drop” form by (1) looking for the presence of pathogens in the basal internodes of stems, (2) quantifying blocked vessels by tylosis and/or gummosis, and (3) describing the impact of the disease on vine physiology (gene expression and metabolome) and development (establishment and functioning of the cambium and phellogen) at the level of xylem and phloem of basal stem internodes. Our study has shown that Botryosphaeriaceae were present in both phloem and xylem of the basal internodes of the annual stem, with xylem vessels obturated. We have also clearly demonstrated that gene expression and metabolite profiles were strongly modified in both xylem and phloem of diseased plants. Differences in stems between healthy (control, C) and diseased (D) plants were low at flowering (vines not yet symptomatic), higher at the onset of symptom expression and still present, although less marked, at full disease expression. qRT-PCR analysis showed in both phloem and xylem an overexpression of genes involved in plant defense, and a repression of genes related to meristematic activity (i.e. vascular cambium and phellogen). Metabolomic analysis showed specific fingerprints in stems of healthy and diseased plants from the onset of symptom expression, with an increase of the level of phytoalexins and mannitol, and a decrease of 1-kestose one. At the structural level, many alterations were observed in internodes, even before the onset of symptoms: a classical obstruction of xylem vessels and, for the first time, a disorganization of the secondary phloem with an obstruction of the sieve plates by callose. The disease modifies the development of both secondary phloem (liber) and phellogen. Altogether, this study combining different approaches allowed to highlight deep vine dysfunction in the internodes at the base of stems, that may explain vine decline due to Botryosphaeria dieback.

Résumé : Abstract Introduction Bleeding severity in severe haemophilic patients, with low thrombin generation (TG) capacity, can vary widely between patients, possibly reflecting differences in tissue factor pathway inhibitor (TFPI) level. Aim To compare free TFPI (fTFPI) levels in patients with severe haemophilia A (sHA) and severe haemophilia B (sHB) and to investigate in these patients as a whole the relationships between bleeding and TG potential, between TG potential and fTFPI level and between fTFPI level and bleeding tendency. Methods Data on bleeding episodes retrospectively recorded during follow‐up visits over 5–10 years were collected and used to calculate the annualised joint bleeding rate (AJBR). fTFPI levels and basal TG parameters were determined in platelet‐poor plasma (PPP) and platelet‐rich plasma (PRP) using calibrated automated tomography (CAT). Results Mean fTFPI levels did not differ significantly between sHA ( n = 34) and sHB ( n = 19) patients. Mean values of endogenous thrombin potential (ETP) and thrombin peak (peak) in PPP and PRP were two‐fold higher when fTFPI levels < 9.4 versus > 14.3 ng/mL. In patients treated on demand, ETP and peak in PRP were doubled when AJBR was , AJBR being halved in patients with a low fTFPI level (9.4 ng/mL). In patients on factor prophylaxis, no association was found between TG parameters and either fTFPI level or AJBR. Conclusion In patients treated on demand, bleeding tendency was influenced by fTFPI levels, which in turn affected basal TG potential. In patients on prophylaxis, bleeding tendency is probably determined primarily by the intensity of this treatment.

Résumé : The French National Committee for Scientific Diving (CNPS) takes on different tasks including acting as an observatory of occupational scientific diving practices and innovations (Thouzeau et al., 2019). Since 1991 in France, scientific diving has been recognized by law as an occupational sector and is therefore regulated with the aim to reduce the hyperbaric risk. Currently, French scientific diving encompasses many fields such as underwater life sciences and ecology, geosciences, social & cultural sciences including archeology, health & food, engineering sciences, energy... Due to the existence of overseas territories - with France's maritime spaces representing the world's second largest exclusive economic zone - French territories display an extensive variety of submarine habitats. Furthermore, the historical richness of scientific diving in France has led the French scientific diving community to be involved in all oceans worldwide. All subaquatic ecosystems, from the polar regions to the tropical belt, from drowned karsts to freshwater and high altitude lakes, including mesocosms and artificial structures are studied by diving scientists. On top of the traditional diving methods (apnea, open circuit and hookah) scientific diving benefited in recent years from the use of closed-circuit rebreathers but also from combining CCR and saturation methods, which greatly enhanced the study of the mesophotic zones. In light of the diversity of environments, disciplines and scientific issues, scientific divers are continuously innovating to perform outstanding research. This presentation will give an overview of recent work from the last decade, highlighting the importance of scientific diving in science in France.

Résumé : Ciliopathies are a class of multi-systemic genetic diseases characterized by ciliary dysfunction. Here, we report a novel ANKS3 variant in patients with a renal ciliopathy known as nephronophthisis (NPH) associated with hepatic defects. ANKS3 is an ankyrin and sterile alpha motif domain-containing protein that interacts with many NPH proteins as well as with BICC1, an RNA-binding protein involved in renal cystic diseases. The pathogenic effect of the ANKS3 mutation was validated in the zebrafish mutant and knock-in rat model, the latter showing urine concentration defect and tubular dilatations similar to NPH patients. In addition, cilia morphology and function as well as epithelialization of kidney tubular cells was affected by loss or mutation of ANKS3 . Finally, our results evidenced that these classically renal ciliopathy-associated phenotypes were linked to the negative regulation of BICC1 by ANKS3 which binds to transcripts of the major NPH gene NPHP1 and mediates their decay through the AGO2-RISC complex and recruitment into P-bodies. Altogether, our findings suggest that the ANKS3/BICC1 complex is a key post-transcriptional regulator of NPHP1 transcript stability, providing another level of regulation of cilium biogenesis and kidney homeostasis, as well as an unusual mechanism leading to NPH-related ciliopathies.

Résumé : It is common that strategic investment decisions are made at a slow time-scale, whereas operational decisions are made at a fast time-scale. Hence, the total number of decision stages may be huge. In this paper, we consider multistage stochastic optimization problems with two time-scales, and we propose a time block decomposition scheme to address them numerically. More precisely, i) we write recursive Bellman-like equations at the slow time-scale and ii), under a suitable monotonicity assumption, we propose computable upper and lower bounds — relying respectively on primal and dual decomposition — for the corresponding slow time-scale Bellman functions. With these functions, we are able to design policies. We assess the methods tractability and validate their efficiency by solving a battery management problem where the fast time-scale operational decisions have an impact on the storage current capacity, hence on the strategic decisions to renew the battery at the slow time-scale.

Résumé : The Coupled Model Intercomparison Project (CMIP) is one of the biggest international efforts aimed at better understanding the past, present, and future of climate changes in a multi-model context. A total of 21 model intercomparison projects (MIPs) were endorsed in its sixth phase (CMIP6), which included 190 different experiments that were used to simulate 40 000 years and produced around 40 PB of data in total. This paper presents the main findings obtained from the CPMIP (the Computational Performance Model Intercomparison Project), a collection of a common set of metrics, specifically designed for assessing climate model performance. These metrics were exclusively collected from the production runs of experiments used in CMIP6 and primarily from institutions within the IS-ENES3 consortium. The document presents the full set of CPMIP metrics per institution and experiment, including a detailed analysis and discussion of each of the measurements. During the analysis, we found a positive correlation between the core hours needed, the complexity of the models, and the resolution used. Likewise, we show that between 5 %–15 % of the execution cost is spent in the coupling between independent components, and it only gets worse by increasing the number of resources. From the data, it is clear that queue times have a great impact on the actual speed achieved and have a huge variability across different institutions, ranging from none to up to 78 % execution overhead. Furthermore, our evaluation shows that the estimated carbon footprint of running such big simulations within the IS-ENES3 consortium is 1692 t of CO2 equivalent. As a result of the collection, we contribute to the creation of a comprehensive database for future community reference, establishing a benchmark for evaluation and facilitating the multi-model, multi-platform comparisons crucial for understanding climate modelling performance. Given the diverse range of applications, configurations, and hardware utilised, further work is required for the standardisation and formulation of general rules. The paper concludes with recommendations for future exercises aimed at addressing the encountered challenges which will facilitate more collections of a similar nature.

Résumé : Spatial genome organization within the nucleus influences major biological processes and is impacted by the configuration of linear chromosomes. Here, we applied 3D spatial statistics and modeling on high-resolution telomere and centromere 3D-structured illumination microscopy images in cancer cells. We found a multi-scale organization of telomeres that dynamically evolved from a mixed clustered-and-regular distribution in early G1 to a purely regular distribution as cells progressed through the cell cycle. In parallel, our analysis revealed two pools of peripheral and internal telomeres, the proportions of which were inverted during the cell cycle. We then conducted a targeted screen using MadID to identify the molecular pathways driving or maintaining telomere anchoring to the nuclear envelope observed in early G1. Lamina-associated polypeptide (LAP) proteins were found transiently localized to telomeres in anaphase, a stage where LAP2α initiates the reformation of the nuclear envelope, and impacted telomere redistribution in the next interphase together with their partner barrier-to-autointegration factor (BAF).
