Résumé : Ce rapport présente des méthodes d’analyse des comportements des cétacés dans des habitats variés et leurs résultats associés, qui seront complétés avec les missions WW5 et les bouées BOMBYX-1,2, et KM3NeT. La surveillance acoustique passive (écoute des vocalises, biosonars ou clics sans interférence avec les cétacés), permet de bien échantillonner les différents groupes d’animaux, individu par individu. Les systèmes de suivi des différents taxons sont maintenant pratiquement opérationnels, seuls la numérisation et le traitement des masses de données bioacoustiques posent encore quelques questions. Pour suivre l’évolution de la biodiversité en milieux ouverts peu accessibles ou peu échantillonnés - ici le vaste volume de l’océan -, les enregistrements sonores sur de larges bandes de fréquence au cours de longues séries temporelles constituent, à ce jour, la meilleure méthode, pour ne pas dire la seule. Les données acoustiques ainsi recueillies permettent de mesurer l’évolution concomitante de l’anthropophonie et des populations de cétacés, même cryptiques.

Les granites syntectoniques sud-armoricains

Résumé : Les géologues structuraux et tectoniciens de Rennes ont non seulement inventé des structures tectoniques remarquables (structure C/S, pli en fourreau), mais ils l'ont fait dans le domaine sud-armoricain, pour certaines d'entre elles à tout le moins. Au-delà des structures mêmes, ils ont posé le cadre tectonique de ce domaine géologique remarquable, où beaucoup de granites d'origine crustale se sont mis en place au Carbonifère, dans les stades avancés de l'orogène varisque. Le cisaillement Sud Armoricain (CSA) est ainsi jalonné de plusieurs granites syn-cinématiques (Pontivy, Lizio), dont la forme cartographique renseigne de la cinématique dextre du cisaillement. Les détachements sud-armoricains ont fonctionné de manière synchrone et sont eux-mêmes le site de mise en place de granites d'origine crustale (Quiberon, Guérande). Les travaux de synthèse de Denis Gapais ont permis de comprendre le fonctionnement général et cohérent de ces structures crustales (CSA et détachements), et ainsi de fournir un cadre aux études pétro-géochimiques qui ont ambitionné de caractériser l'évolution pétrogénétique même des granites, et d'intégrer l'histoire des circulations de fluides associées. La reconnaissance de l'origine météorique de certains composants de ces fluides a permis d'initier la reconstitution des paléoreliefs de la zone au Carbonifère, et d'éclairer les modalités de mise en place de certains systèmes métallogéniques, notamment uranifères (thèses de Romain Tartèse et Christophe Ballouard, soutenues naguère à l'Université de Rennes 1). Aujourd'hui, les travaux de caractérisation pétro-géochimique toujours plus fine des interactions entre fluides, granites et zones de déformations sont enrichis d'une modélisation numérique hydro-thermale, qui apporte des contraintes physiques au fonctionnement des systèmes (thèse de Khaled Bock, en préparation à l'Université de Rennes). Dans cette présentation, je reprendrai quelques-uns des jalons principaux de cette histoire granitique armoricaine, dont les titres de chapitre doivent beaucoup aux tectoniciens de Rennes, Pierre Choukroune et Jean-Pierre Brun en tête, mais aussi Denis Gapais et Peter Cobbold, en passant par Pierre Jégouzo, le Morbihannais de l'étape, et Claude Le Corre.

Résumé : Objective: To evaluate the efficacy of covid-19 convalescent plasma to treat patients admitted to hospital for moderate covid-19 disease with or without underlying immunodeficiency (CORIPLASM trial). Design: Open label, randomised clinical trial. Setting: CORIMUNO-19 cohort (publicly supported platform of open label, randomised controlled trials of immune modulatory drugs in patients admitted to hospital with moderate or severe covid-19 disease) based on 19 university and general hospitals across France, from 16 April 2020 to 21 April 2021. Participants: 120 adults (n=60 in the covid-19 convalescent plasma group, n=60 in the usual care group) admitted to hospital with a positive SARS-CoV2 test result, duration of symptoms <9 days, and World Health Organization score of 4 or 5. 49 patients (n=22, n=27) had underlying immunosuppression. Interventions: Open label randomisation to usual care or four units (200-220 mL/unit, 2 units/day over two consecutive days) of covid-19 convalescent plasma with a seroneutralisation titre >40. Main outcome measures: Primary outcomes were proportion of patients with a WHO Clinical Progression Scale score of ≥6 on the 10 point scale on day 4 (higher values indicate a worse outcome), and survival without assisted ventilation or additional immunomodulatory treatment by day 14. Secondary outcomes were changes in WHO Clinical Progression Scale scores, overall survival, time to discharge, and time to end of dependence on oxygen supply. Predefined subgroups analyses included immunosuppression status, duration of symptoms before randomisation, and use of steroids. Results: 120 patients were recruited and assigned to covid-19 convalescent plasma (n=60) or usual care (n=60), including 22 (covid-19 convalescent plasma) and 27 (usual care) patients who were immunocompromised. 13 (22%) patients who received convalescent plasma had a WHO Clinical Progression Scale score of ≥6 at day 4 versus eight (13%) patients who received usual care (adjusted odds ratio 1.88, 95% credible interval 0.71 to 5.24). By day 14, 19 (31.6%) patients in the convalescent plasma group and 20 (33.3%) patients in the usual care group needed ventilation, additional immunomodulatory treatment, or had died. For cumulative incidence of death, three (5%) patients in the convalescent plasma group and eight (13%) in the usual care group died by day 14 (adjusted hazard ratio 0.40, 95% confidence interval 0.10 to 1.53), and seven (12%) patients in the convalescent plasma group and 12 (20%) in the usual care group by day 28 (adjusted hazard ratio 0.51, 0.20 to 1.32). In a subgroup analysis performed in patients who were immunocompromised, transfusion of covid-19 convalescent plasma was associated with mortality (hazard ratio 0.39, 95% confidence interval 0.14 to 1.10). Conclusions: In this study, covid-19 convalescent plasma did not improve early outcomes in patients with moderate covid-19 disease. The efficacy of convalescent plasma in patients who are immunocompromised should be investigated further.

Résumé : Owing to its capability to produce large volume of glass while preserving a high degree of purity and homogeneity, the suspension method was employed here to synthesize a Yb 3+ /Al 3+ /P 5+ doped silica preforms. The glass structure was studied by relying on both nuclear magnetic resonance (NMR) and Raman spectroscopies, confirming the formation of Al(PO) 4 units. Thence, photoluminescence emission spectra were acquired, assessing the beneficial impact of the phosphorus addition in Yb/Al doped silica glass to curtail the Yb 2+ ions content. The results reported here suggest that alumino - phosphosilicate matrix having an equimolar concentration of Al 3+ /P 5+ co-dopants exhibits significantly weaker concentration in Yb 2+ ions than equivalent aluminosilicate matrix. This glass composition is thus shown relevant to look further on circumventing the photodarkening phenomenon occurring into fiber laser.

Résumé : In routine hematological instruments, blood cells are counted and sized by monitoring the impedance signals induced during their passage through a Coulter orifice. However, only signals associated with centered paths in the aperture are considered for analysis, while the rejected measurements, caused by near-wall trajectories, can provide additional information on red blood cells (RBC), as recent publications suggest. To assess usefulness of two new parameters in describing alterations in RBC properties, we performed a pilot study to compare blood samples from patients with diabetes mellitus (DM), frequent pathological condition associated with impairment in RBC deformability, versus controls. A total of 345 blood samples were analyzed: 225 in the DM group and 120 in the control group. A diagram of R and P , the two new parameters derived from the analysis of impedancemetry pulses, was used to compare distribution of RBC subpopulations between groups. To discriminate RBC from DM and control individuals, based on our multiparametric analysis, we built a score from variables derived from R / P matrix which showed good performances: area under the receiving operating characteristic curve 0.948 (0.920–0.970), p<0.0001; best discriminating value: negative predictive value 94.7%, positive predictive value was 78.4%. These results seem promising to approach RBC alterations in routine laboratory practice. The related potential clinically relevant outcomes remain to be investigated.

Résumé : The classical Non-Homologous End Joining (c-NHEJ) pathway is the predominant process in mammals for repairing endogenous, accidental or programmed DNA Double-Strand Breaks. c-NHEJ is regulated by several accessory factors, post-translational modifications, endogenous chemical agents and metabolites. The metabolite inositol-hexaphosphate (IP6) stimulates c-NHEJ by interacting with the Ku70–Ku80 heterodimer (Ku). We report cryo-EM structures of apo- and DNA-bound Ku in complex with IP6, at 3.5 Å and 2.74 Å resolutions respectively, and an X-ray crystallography structure of a Ku in complex with DNA and IP6 at 3.7 Å. The Ku-IP6 interaction is mediated predominantly via salt bridges at the interface of the Ku70 and Ku80 subunits. This interaction is distant from the DNA, DNA-PKcs, APLF and PAXX binding sites and in close proximity to XLF binding site. Biophysical experiments show that IP6 binding increases the thermal stability of Ku by 2°C in a DNA-dependent manner, stabilizes Ku on DNA and enhances XLF affinity for Ku. In cells, selected mutagenesis of the IP6 binding pocket reduces both Ku accrual at damaged sites and XLF enrolment in the NHEJ complex, which translate into a lower end-joining efficiency. Thus, this study defines the molecular bases of the IP6 metabolite stimulatory effect on the c-NHEJ repair activity.

Résumé : Background: In MAPT (Multidomain Alzheimer Preventive Trial), a cognitive effect of multidomain intervention (MI) was showed in non-demented subjects with positive amyloid PET. However, screening eligible patients for multidomain intervention by PET is difficult to generalize in real-world settings. Methods: MAPT study was a 3-year, randomized, placebo-controlled trial followed by a 2-year observational and optional extension. All participants were non-demented and randomly assigned (1:1:1:1) to the MI plus omega 3, MI plus placebo, omega 3 alone, or placebo alone group. The objectives were to assess the cognitive effect of MAPT interventions (omega 3 supplementation, MI, combined intervention) in non-demented subjects according to amyloid blood status at 12, 36, and 60 months. In this subgroup analysis (n = 483), amyloid status was defined by plasma Aβ42/40 ratio (cutoff ≤ 0.0107). The primary outcome measure was the change in cognitive composite score after a 1, 3, and 5-year clinical follow-up. Results: The intention-to-treat (ITT) population included 483 subjects (161 positive and 322 negative amyloid participants based on plasma Aβ42/40 ratio). In the positive amyloid ITT population, we showed a positive effect of MI plus omega 3 on the change in composite cognitive score in 12 (raw p = .0350, 0.01917, 95% CI = [0.0136 to 0.3699]) and 36 months (raw p = .0357, 0.2818, 95% CI = [0.0190 to 0.5446]). After correction of multiple comparisons and adjustments, these differences were not significant (adjusted p = .1144 and .0690). In the per-protocol positive amyloid group (n = 154), we observed a significant difference between the combined intervention and placebo groups at 12 (p = .0313, 0.2424, 0.0571 to 0.4276) and 36 months (p = .0195, 0.3747, 0.1055 to 0.6439) persisting after adjustment. In the ITT and per-protocol analyses, no cognitive effect was observed in the positive and negative amyloid group at 60-month visit. Conclusions: These findings suggest a benefit of MI plus omega 3 in positive blood amyloid subjects. This promising trend needs to be confirmed before using blood biomarkers for screening in preventive trials. Trial registration: ClinicalTrials.gov Identifier: NCT01513252 .

Résumé : The seabed of the English Channel (EC) corresponds to an extensive network of paleo-valleys, partly filled at the present-day with coarse sediments, reaching sometimes several metres thick, which represent an important source of accessible aggregates. The extraction of these aggregates is currently experiencing significant growth along the French coast of the EC. Despite coarse sediments constitutes approximately 80% of the superficial composition of the EC, very few quantitative studies have been conducted on the benthic community which colonise these habitats. This lack of quantitative data and studies is primarily attributed to the challenges associated with sampling in this area, mainly due to its hydrodynamic conditions. In this context, the aim of this study was then to analyse the spatio-temporal evolution of benthic communities in relation to aggregate extraction. To achieve this, a Before-After Control-Impact (BACI) approach was employed. Thus, samples were collected before extraction between 2007 and 2010, then, two samplings were made during aggregate extraction in 2018 and 2023. The extraction activity was limited to a single strip within the concession, responsible of the refinement of the coarse sand. In 2023, two video acquisitions were carried out to assess the effects of marine aggregate extraction on the benthic community. Although the benthic diversity on the site appeared similar in 2023, a notable decrease in benthic abundance was observed. Techniques investigating the sediment-benthos relationship have also been applied to determine whether sediment reworking has had an impact on “grain size-sensitive” species, particularly those preferring coarse substrates.

Résumé : Abstract Objectives Limited pharmacokinetics data support dalbavancin long-term use in off-label indications and the optimal dosing regimen is debated. We aimed to describe dalbavancin concentrations in an observational retrospective multicentre study. Methods Patients from 13 French hospitals, treated with 1500 mg doses of dalbavancin and for whom therapeutic drug monitoring was performed from June 2018 to March 2021 were included. Dalbavancin plasma concentrations were described at peak and 1, 2, 3, 4, 6 and 8 weeks after the last 1500 mg dose. Concentrations in patients weighing more or less than 75 kg and with a GFR greater or less than 60 mL/min were compared. Microbiological data were collected and dalbavancin MIC was measured when possible. Results One hundred and thirty-three patients were included (69% treated for bone and joint infections, 16% for endocarditis). Thirty-five patients received a single dose of dalbavancin and 98 received several administrations. Two, 3 and 4 weeks after the last dose, median plasma concentrations were respectively 25.00, 14.80 and 9.24 mg/L for the first doses and 34.55, 22.60 and 19.20 mg/L for the second or subsequent doses. Weight and renal function had an impact on pharmacokinetics. Infection was documented in 105 patients (Staphylococcus spp. in 68% of cases). Staphylococcus aureus was isolated in 32.5% of cases (median MIC: 0.047 mg/L) and Staphylococcus epidermidis in 27% of cases (median MIC of 0.047 mg/L). Conclusions Plasma concentrations of dalbavancin were consistent with those described in clinical trials and those sought during the industrial development of the molecule.
