Résumé : IMPORTANCE Extended-spectrum β-lactamase (ESBL)-producing Escherichia coli is considered a leading pathogen contributing to the global burden of antimicrobial resistance. OBJECTIVE To better understand factors associated with the heterogeneity of community-acquired ESBL-producing E coli urinary tract infections (UTIs) in France. DESIGN, SETTING, AND PARTICIPANTS This cross-sectional study performed from January 1 to December 31, 2021, was based on data collected via PRIMO (Surveillance and Prevention of Antimicrobial Resistance in Primary Care and Nursing Homes), a nationwide clinical laboratory surveillance system in France. Strains of E coli isolated from community urine samples from January 1 to December 31, 2019, from 59 administrative departments of metropolitan France were included. MAIN OUTCOMES AND MEASURES Quasi-Poisson regression models were used to assess the associations between several ecological factors available on government and administration websites between 2010 and 2020 (demographic population structure, living conditions, baseline health care services, antibiotic consumptions, economic indicators, animal farming density, and environmental characteristics) and the number of ESBL-producing E coli strains isolated from urine samples of individuals with community-acquired UTI in 2019.

Résumé : In the context of underground exploitation, the behaviour of rocks near galleries and tunnels conditions their stability. Underground drilling generates deformations, damage, fracturing, and significant modification of flow characteristics in the surrounding rock. However, the influence of small-scale characteristics and behaviour on the rock deformations and damage at engineering scale remains a complex issue. Consequently, the multiscale behaviour of a clay rock is modelled starting from the large scale of the excavation damaged zone around galleries and then enriching the approach by considering microstructural characteristics from the scale of mineral inclusions. Lastly, a double-scale numerical framework is considered. It allows to relate small- to large-scale rock behaviour in terms of deformations and material rupture. In fact, the development of damage and cracking at microscale allows to predict large-scale fracturing. The developed method focuses on a claystone in the particular context of long-term management of high-level nuclear wastes by deep geological repository. The results highlight the possibilities of double-scale computing in the prediction of the behaviour of underground engineering structures.

Résumé : Background: Deregulation of energy balance during obesity is associated with the development of metabolic diseases such as type 2 diabetes (T2D). Genetic association studies have established the tumor suppressor locus CDKN2A (cyclin dependent kinase inhibitor 2a) as a risk factor associated with T2D. Consistently, CDKN2A expression in adipose tissue of obese patients is increased. Overall, this locus has a dual function in the regulation of the cell cycle and energy metabolism. We previously showed that Cdkn2a-/- mice exhibit increased energy expenditure via browning of inguinal white adipose tissue (WAT) and improved insulin sensitivity following obesity induced by a high-fat diet (DIO). Goal of the study: To assess whether beiging is cell-autonomously controlled by Cdkn2a and due to transdifferentiation of mature adipocyte and/or recruitment of adipocyte precursors, we analyzed the effect of Cdkn2a deficiency restricted to mature adipocytes in mice (Cdkn2aad-/-). Material et methods: Metabolic studies were performed on mice under chow diet and DIO. Metabolic, hormonal, histological parameters as well as glucose homeostasis were assessed. Histological studies of adipose tissues were also performed. The adipose tissue transcriptome was analyzed by RNA-sequencing. Results: Although metabolic parameters were comparable between controls and mutant animals in chow diet, metabolic, hormonal, histological parameters as well as glucose homeostasis were different in Cdkn2aad-/- DIO mice compared to control littermates. Despite normal food intake, body and fat pad weights, Cdkn2aad-/- DIO mice displayed an improved metabolic profile associated to increased energy expenditure (p<0.01) and improved glucose tolerance (p<0.05) without any changes in insulin sensitivity. Mice also exhibited reduced circulating levels of leptin and resistin (p<0.05). Histological analysis revealed epididymal WAT hypertrophy and inguinal WAT hypoplasia (p<0.05) without effects on proliferation or browning. Transcriptomic analysis of epididymal WAT showed a global increase in metabolic pathways involved in energy substrate utilization, especially glucose metabolism. In particular, a decrease in p70S6K signaling pathway (-log(p-value) 5.540), which is known to reduce insulin signaling, suggests a better use of glucose in Cdkn2a-deficient adipocytes. Conclusion: In summary, these results suggest that the deletion of Cdkn2a in fully differentiated adipocytes is not sufficient to induced WAT browning. Our data provide evidence that the Cdkn2a locus plays a key role in energy metabolism in mature adipocytes, making this gene a good candidate to alleviate the effects of obesity and prevent insulin resistance and T2D.

Résumé : The “gel casting” is a process used for producing parts with complex geometries. It was initially developed to overcome the limitations of other shaping processes such as injection moulding or machining. Designed by Janney & Omatete [1] in 1991, this process is very versatile, both in terms of its compatibility with different classes of materials (ceramics and metals) and its compatibility with different mould manufacturing techniques. In practice, gel casting is based on the production of a slurry in an aqueous medium in the presence of a dispersant and a gelling agent. The suspension is pouring into a mould where the gelling can occur and form a three-dimensional network which ensures the conservation of the different parameters of the slurry. The desired part is obtained after drying and then demolding while a debinding/sintering heat treatment ensures its densification. Although the approach is productive and efficient for high quality complexes object, but is it still limited by the need for a manufacturing technic for the mould. Its use is also limited by the toxicity of polymers gelling systems based on acrylamide. The idea for this project is therefore to work on an indirect additive manufacturing process combining stereolithography (SLA) polymer and gel casting. Additive manufacturing allows the manufacture of casting moulds while gel casting allows the shaping of ceramic parts. The coupling of these two techniques theoretically allows the manufacture of parts with very complex shape where only the ability to unmould and dry the sample is a hurdle. Résumé : Objective To quantify the risk of immune-related adverse events (irAEs) in patients with pre-existing autoimmune disease (pAID) treated by immune checkpoint inhibitors (ICIs) for stage III or IV melanoma. Methods Case–control study performed on a French multicentric prospective cohort of patients with melanoma, matched for irAE risk factors and oncological staging. Risk of irAE was assessed by logistic regression. Results 110 patients with pAID were included and matched with 330 controls, from March 2013 to October 2020. Over a median follow-up period of 7.2 months for cases and 6.9 months for controls, the ORs of developing all-grade and grade ≥3 irAEs among cases compared with controls were 1.91 (95% CI (1.56 to 2.27)) and 1.44 (95% CI (1.08 to 1.82)), respectively. Patients with pAID had an increased risk of multiple irAEs (OR 1.46, 95% CI (1.15 to 2.67)) and a shorter time to irAE onset. In contrast, there were no difference in irAE-related mortality nor in the rate of treatment discontinuation, and a landmark analysis revealed a better survival at 24 months among cases (p=0.02). Thirty per cent of cases experienced a pAID flare during follow-up, and baseline immunosuppression did not prevent irAE occurrence. Last, we report associations between the pAID clinical subsets and organ-specific irAEs. Conclusion In our study, patients with pAID were at greater risk of all-grade, severe and multiple irAEs, yet had a better 24-month survival than controls. Thus, patients with pAID should be eligible for ICI therapy but benefit from a close monitoring for irAE occurrence, especially during the first months of therapy.

Résumé : This chapter addresses collection and integration of large amounts of data from a variety of sources when developing agronomic approaches at the landscape and territory levels. Data commonly used in agro-environmental studies are climate, soils, land cover and land use (including cropping practices). They are used as inputs for indicator- or model-based assessment methods. Additional information is now required for agroecological studies, such as spatial distribution of weeds, pests, and diseases; biodiversity; or landscape features. Gathering and using this scattered and heterogeneous information for integrated studies at watershed, regional, or national levels still requires further methodological efforts. Remote sensing is a source of data that continuously progresses in spatial and temporal resolution, and accessibility. Such integrative approaches are illustrated for a case study in France. The next challenges and opportunities for data collection, integration, and governance are discussed, with a focus on mainland France.
