Résumé : Background: It is not clear whether pre-existing macrovascular complications (ischemic heart disease, stroke or peripheral artery disease) are associated with health outcomes in people with diabetes mellitus hospitalized for COVID-19. Methods: We conducted cohort studies of adults with pre-existing diabetes hospitalized for COVID-19 infection in the UK, France, and Spain during the early phase of the pandemic (between March 2020-October 2020). Logistic regression models adjusted for demographic factors and other comorbidities were used to determine associations between previous macrovascular disease and relevant clinical outcomes: mortality, intensive care unit (ICU) admission and use of invasive mechanical ventilation (IMV) during the hospitalization. Output from individual logistic regression models for each cohort was combined in a meta-analysis. Results: Complete data were available for 4,106 (60.4%) individuals. Of these, 1,652 (40.2%) had any prior macrovascular disease of whom 28.5% of patients died. Mortality was higher for people with compared to those without previous macrovascular disease (37.7% vs 22.4%). The combined crude odds ratio (OR) for previous macrovascular disease and mortality for all four cohorts was 2.12 (95% CI 1.83-2.45 with an I2 of 60%, reduced after adjustments for age, sex, type of diabetes, hypertension, microvascular disease, ethnicity, and BMI to adjusted OR 1.53 [95% CI 1.29-1.81]) for the three cohorts. Further analysis revealed that ischemic heart disease and cerebrovascular disease were the main contributors of adverse outcomes. However, proportions of people admitted to ICU (adjOR 0.48 [95% CI 0.31-0.75], I2 60%) and the use of IMV during hospitalization (adjOR 0.52 [95% CI 0.40-0.68], I2 37%) were significantly lower for people with previous macrovascular disease. Conclusions: This large multinational study of people with diabetes mellitus hospitalized for COVID-19 demonstrates that previous macrovascular disease is associated with higher mortality and lower proportions admitted to ICU and treated with IMV during hospitalization suggesting selective admission criteria. Our findings highlight the importance correctly assess the prognosis and intensive monitoring in this high-risk group of patients and emphasize the need to design specific public health programs aimed to prevent SARS-CoV-2 infection in this subgroup.

Résumé : Our objective was to explore chemical strategies based on organometallic precursors in organic solvents to yield unprecedented TiO2- or ZnO-decorated SBA-15. We synthesized hybrid materials (TiO2/SBA-15 or ZnO/SBA-15) from various organometallic precursors such as tetrakis(ethylmethylamido) titanium (TEMAT), titanium tris-amidinate (Ti-Amd) and dicyclohexyl zinc (Zn(Cy)2), and under various hydration conditions. In future work, the efficiency of such materials will be assessed under injection of 400 ppb NO.

Résumé : Since the acceptance of plate tectonics, the presence of calc-alkaline magmatic rocks has been recognized as evidence of subduction. But under specific geodynamic circumstances, subduction may occur without generating magmas. Here, we investigate the Cenozoic northern Lesser Antilles arc where, from sparsely exposed magmatic records, Eocene−Oligocene and Pliocene magmatic flare-ups and a Miocene lull were postulated. Nevertheless, most of the arc is submarine, so it is challenging to discern lulls and flare-ups from sampling bias. We review the magmatic evidence exposed onshore in the Lesser Antilles and investigate in detail the island of Antigua, which exposes an Eocene to Miocene volcanic sequence and platform carbonate series that coincide with the postulated lull. By combining lithostratigraphic analysis, structural mapping, 40Ar/39Ar geochronology, and biostratigraphy, we refine the magmatic history of the island and date the arrest of extensive arc magmatism at 35 Ma, with minor activity until 27 Ma. No magmatic products are interleaved with the platform sequence until the latest Oligocene, which confirms a lull in northern Lesser Antilles arc magmatism that may have lasted ca. 20 Ma. Flare-up of magmatic activity contributed to crustal thickening and land emersion, whereas magmatic lulls and related thermal cooling induced subsidence/submersion. Thus, we propose that the paleo-(bio)-geographical evolution of the eastern Caribbean region has been partly controlled by magmatic activity. Fault kinematic analysis, along with anisotropy of magnetic susceptibility, suggest that, at the island scale, magmatic arrest is not associated with a change in stress field during the Oligocene. We speculate that slab flattening triggered by progressive curvature played a role in the temporal shutdown of the northern Lesser Antilles arc.

Résumé : L’Annuaire de Justice transitionnelle entend contribuer à susciter et à diffuser une pensée francophone sur la justice transitionnelle, comprise comme incluant divers thèmes allant de la justice pénale (nationale ou internationale) aux commissions vérité et réconciliation, en passant par toute une série de mesures pouvant être adoptées à l’issue d’un conflit armé ou après la chute d’un régime autoritaire. Dans ces hypothèses de retour à la paix ou de transition démocratique (qui ne sont, du reste, pas toujours pérennes), il s’agit d’analyser les réponses offertes par le droit et par d’autres sciences humaines pour que des sociétés meurtries affrontent leur passé et préparent leur avenir dans les meilleures conditions. Outre une ouverture internationale et pluridisciplinaire assumée, la nouvelle mouture de l’Annuaire vise à publier des contributions écrites par des universitaires mais aussi des praticiens, jeunes ou confirmés, qu’il s’agisse de mémoires de recherche réalisés par des étudiants en master 2, d’actes de colloques académiques, de témoignages de praticiens de la justice pénale ou transitionnelle ou encore d’études sur des expériences passées ou à venir, avec une perspective comparatiste dans le temps et dans l’espace. Ces articles sont utilement complétés d’une bibliographie francophone de l’année en cours, comprenant, outre la doctrine pertinente, les références des principales jurisprudences internationales et nationales et une liste des événements marquants de la justice pénale internationale et transitionnelle. L’Annuaire 2021 comprend notamment les actes de deux colloques universitaires, le rapport de synthèse du Séminaire annuel de l’IFJD, le dossier spécial « Processus de paix », des contributions variées, ainsi que la chronique de l’année.

Résumé : Cerebral malaria (CM) is one of the most severe forms of malaria and is a neuropathology that can lead to death. Monocytes have been shown to accumulate in the brain microvasculature at the onset of neurological symptoms during CM. Monocytes have a remarkable ability to adapt their function to their microenvironment from pro-inflammatory to resolving activities. This study aimed to describe the behavior of monocyte subpopulations during infection and its resolution. C57BL/6 mice were infected with the Plasmodium berghei ANKA strain and treated or not with chloroquine (CQ) on the first day of the onset of neurological symptoms (day 6) for 4 days and followed until day 12 to mimic neuroinflammation and its resolution during experimental CM. Ly6C monocyte subpopulations were identified by flow cytometry of cells from the spleen, peripheral blood, and brain and then quantified and characterized at different time points. In the brain, the Ly6C int and Ly6C low monocytes were associated with neuroinflammation, while Ly6C hi and Ly6C int were mobilized from the peripheral blood to the brain for resolution. During neuroinflammation, CD36 and CD163 were both involved via splenic monocytes, whereas our results suggest that the low CD36 expression in the brain during the neuroinflammation phase was due to degradation. The resolution phase was characterized by increased expressions of CD36 and CD163 in blood Ly6C low monocytes, a higher expression of CD36 in the microglia, and restored high expression levels of CD163 in Ly6C hi monocytes localized in the brain. Thus, our results suggest that increasing the expressions of CD36 and CD163 specifically in the brain during the neuroinflammatory phase contributes to its resolution.

Résumé : Keratoconus (KC) is a multifactorial progressive ectatic disorder characterized by local thinning of the cornea, leading to decreased visual acuity due to irregular astigmatism and opacities. Despite the evolution of advanced imaging methods, the exact etiology of KC remains unknown. Our aim was to investigate the involvement of corneal epithelium in the pathophysiology of the disease. Corneal epithelial samples were collected from 23 controls and from 2 cohorts of patients with KC: 22 undergoing corneal crosslinking (early KC) and 6 patients before penetrating keratoplasty (advanced KC). The expression of genes involved in the epidermal terminal differentiation program and of the oxidative stress pathway was assessed by real time PCR analysis. Presence of some of the differentially expressed transcripts was confirmed at protein level using immunofluorescence on controls and advanced KC additional corneal samples. We found statistically significant under-expression in early KC samples of some genes known to be involved in the mechanical resistance of the epidermis ( KRT 16, KRT14 , SPRR1A , SPRR2A , SPRR3 , TGM1 and TGM5 ) and in oxidative stress pathways ( NRF2 , HMOX1 and HMOX2 ), as compared to controls. In advanced KC samples, expression of SPRR2A and HMOX1 was reduced. Decreased expression of keratin (KRT)16 and KRT14 proteins was observed. Moreover, differential localization was noted for involucrin, another protein involved in the epidermis mechanical properties. Finally, we observed an immunofluorescence staining for the active form of NRF2 in control epithelia that was reduced in KC epithelia. These results suggest a defect in the mechanical resistance and the oxidative stress defense possibly mediated via the NRF2 pathway in the corneal keratoconic epithelium.

Résumé : Symbiotic interactions have structured past and present ecosystems and shaped the evolution of life. As any trait, the symbiotic state observed in extant species builds on ancestral and conserved features, and lineage-specific innovations. From these mixed origins, defining the ancestral state of symbiotic associations is challenging although it is instrumental for understanding how symbiotic abilities emerge from non-symbiotic states. Here we aimed at reconstructing the intermediate steps leading to the root-nodule nitrogen-fixing symbiosis (RNS) observed in some extant flowering plants. For this, we compared the transcriptomic responses of nine host plants in response to symbiotic bacteria. We included the mimosoid legume Mimosa pudica for which we assembled a chromosome-level genome and generated the transcriptomic response to experimentally evolved bacterial symbionts. With this dataset, we reconstructed the ancestral RNS transcriptome, composed of most already described symbiotic genes together with hundreds of novel candidates. We found that the response to the chemical signals produced by the symbiont, nodule organogenesis and nitrogen-fixation are predominantly linked to ancestral responses, although these traits have diversified in the different nitrogen-fixing lineages. We detected a clear signature of recent and convergent evolution for the ability to release intracellular symbiosomes in two legume lineages, exemplified by the expression of different classes of small proteins in each group, potentially leading to the convergent gain of symbiotic evolutionary stability. Our findings demonstrate that most of the novelties for RNS were mostly in place in the most recent common ancestor of the RNS-forming species that lived on Earth 110 million years ago.
