Résumé : Gaucher Disease (GD) is a rare genetic disorder characterized by a deficiency in the enzyme glucocerebrosidase, leading to the accumulation of glucosylceramide in various cells, including red blood cells (RBCs). This accumulation results in altered biomechanical properties and rheological behavior of RBCs, which may play an important role in blood rheology and the development of bone infarcts, avascular necrosis (AVN) and other bone diseases associated with GD. In this study, dissipative particle dynamics (DPD) simulations are employed to investigate the biomechanics and rheology of blood and RBCs in GD under various flow conditions. The model incorporates the unique characteristics of GD RBCs, such as decreased deformability and increased aggregation properties, and aims to capture the resulting changes in RBC biophysics and blood viscosity. This study is the first to explore the Young's modulus and aggregation parameters of GD RBCs by validating simulations with confocal imaging and experimental RBC disaggregation thresholds. Through in silico simulations, we examine the impact of hematocrit, RBC disaggregation threshold, and cell stiffness on blood viscosity in GD. The results reveal three distinct domains of GD blood viscosity based on shear rate: the aggregation domain, where the RBC disaggregation threshold predominantly influences blood viscosity; the transition area, where both RBC aggregation and stiffness impact on blood viscosity; and the stiffness domain, where the stiffness of RBCs emerges as the primary determinant of blood viscosity. By analyzing RBC mechanical properties and blood viscosity in relation to bone disease, we find that the RBC aggregation properties, deformability, and blood viscosity, may contribute to its onset. These findings enhance our understanding of how changes in RBC properties impact on blood viscosity and may affect bone health, offering a partial explanation for the bone complications observed in GD patients.

Résumé : Abstract The lateral prefrontal cortex (LPFC) serves as a critical hub for higher-order cognitive and executive functions in the human brain, coordinating brain networks whose disruption has been implicated in many neurological and psychiatric disorders. While transcranial brain stimulation treatments often target the LPFC, our current understanding of connectivity profiles guiding these interventions based on electrophysiology remains limited. Here, we present a high-resolution probabilistic map of bidirectional effective connectivity between the LPFC and widespread cortical and subcortical regions. This map is derived from intracranial evoked potential analysis of 48,797 intracranial direct electrical stimulation runs across 759 implantations in 724 patients with refractory epilepsy (368 male, 354 female, two unspecified; mean age 24±13.5 years). We mapped probabilistic connectivity between brain parcels with adaptive resolution – higher resolution in the LPFC in the hemisphere of interest and lower elsewhere – maintaining statistical power while achieving 95% average confidence interval of ∼0.03 for connectivity probability estimates. In addition, the significance threshold (p-value) for probabilistic connectivity was obtained from surrogate distributions. Overall, we observed remarkable symmetry between afferent and efferent connectivity patterns of the LPFC, with a slight preference for efferent connections (mean slope = 0.92±0.09, mean R² = 0.93±0.025). For example, connections between the inferior frontal gyrus (IFG) and anterior cingulate showed notable directional asymmetry. The IFG strongly projected to most brain networks compared to other LPFC regions, with the strongest connectivity to the ventral attention network (0.26±0.01 compared to values between 0.15 and 0.21 in other LPFC regions). Posterior DLPFC demonstrated stronger connectivity to brain networks compared to anterior DLPFC regions (eg. 0.21±0.01 vs 0.15±0.01 for connectivity to ventral attention network), with the exception of the limbic cortex. All LPFC subregions strongly projected to the fronto-parietal (greater than 0.17) and ventral attention (greater than 0.15) networks, with moderate connections to the default network (between 0.1 and 0.15, with the maximum corresponding to superior DLPFC). Finally, latency analysis suggested that the left LPFC's influence on ipsilateral emotion-related regions is primarily polysynaptic, with particularly strong pathways from IFG to amygdala (0.16±0.02) and hippocampus (0.12±0.01). Taken together, these comprehensive connectivity maps provide a new detailed electrophysiological foundation for understanding the functional anatomy of LPFC and guiding targeted brain stimulation protocols.

Résumé : Abstract. A comprehensive meteorological data set from an operational wind farm, consisting of six 2 MW turbines, has been made available. A meteorological mast, equipped with sonic anemometers at four different heights, was installed at the center of the farm and has collected data over 3 years. The data set is further supplemented with radiometer measurements for atmospheric stability analysis. Simultaneously, supervisory control and data acquisition (SCADA) data were acquired to provide operational information about the wind turbines, including inter alia power production and wind direction. Additionally, the turbine blades were scanned to support aerodynamic simulations. This unique and comprehensive database has been made accessible to the research community through the AERIS platform.

Résumé : Salt crystallization within the heritage stones is recognized as a major cause of damage. In particular, sodium sulfates which are very commonly encountered in natural environment, are recognized as the most destructive salts. The mechanisms leading to the stone damage are still not well defined. The identification of the salt crystallization sequence and of the spatial distribution at the pore scale, remain a real scientific challenge. The sodium sulfate crystallization within true natural porous network is studied at pore scales during weathering cycles. Savonnières limestone samples are exposed to salt by successive imbibition and evaporation of a 1 mol/L sodium sulfate solution at constant ambient temperature and relative humidity conditions. Phases characterization and topography cartographies are performed using Raman spectroscopy and Atomic Force Microscopy respectively. The two combined technics on a same limestone sample help to characterize the spatial distribution of neocrystallized salt phases within the porous network. The results shown that the micro and the macropores of the limestones are mainly filled by thenardite phase V microcrystals. Large crystals of mirabilite could be formed in the stone before the dehydration. Gypsum crystals have been observed too and are located in the micropores of the limestone: at interfacial zones between oolites and sparitic cement and also in the micropores of the oolites. The presence of gypsum could indicate a chemical reaction between the sodium sulfate solution and the limestone.

Résumé : Background and aims Inflammatory bowel disease (IBD), including Crohn’s disease (CD) and ulcerative colitis (UC), remain heterogeneous disorders with variable response to biologics. Post-operative recurrence in CD is common despite surgery and prophylactic biotherapies. Understanding the inflammatory mediators associated with recurrence and treatment response could pave the way for personalized strategies. Methods We analyzed serum inflammatory protein signatures using proteomics in two prospective cohorts. The REMIND cohort included post-operative CD patients undergoing ileocecal resection with endoscopic assessment at 6 months (M6). Serum samples were collected at surgery and six months later. The ELYP cohort consisted of active IBD patients starting new biotherapies (anti-TNF, ustekinumab, or vedolizumab). Serum samples were collected pre- and post-treatment (Weeks 14 and 52). Results In the REMIND cohort, proteomic analysis revealed elevated levels of IFN-γ, CXCL9, and MMP-10 in patients with recurrence, with concentrations associated with recurrence severity. Preoperative MMP-10 levels predicted severe recurrence (AUC = 0.70). Under biotherapies, treatment-specific proteins were associated with recurrence: CXCL9 for anti-TNF and OSM/TGFα modules for ustekinumab. In the ELYP cohort, IFN-γ and CXCL9 were significantly elevated in CD compared to UC and associated with disease activity. Early response to anti-TNF treatment (Week 14) was associated with reductions in CXCL9, MMP-10, and OSM, while deep remission (Week 52) correlated with decreases in CXCL9 and OSM. Conclusion Our findings reveal inflammatory blood proteomic signatures associated with post-operative recurrence and biologic treatment failure in IBD. Several key biomarkers were identified. Résumé : The anisotropic flow coefficients $v_n$ quantify the collective medium response to the initial spatial anisotropy of the overlapping region in ion collisions and serve as sensitive probes of both the medium properties and shape of nuclear initial states. In this analysis, the $v_2$ and $v_3$ parameters of prompt charged particles are measured using the multiparticle cumulant method in fixed-target PbNe and PbAr collisions at $\sqrt{s_{NN}} = 70.9$ GeV, collected by LHCb using the SMOG2 gas-target system during the 2024 LHC lead-beam run. The cumulant method is first validated using 2018 PbPb collision data, successfully reproducing previous measurements obtained via the two-particle correlation method. Results for the fixed-target collisions are then presented, showing a significantly larger value of the elliptic flow coefficient $v_2$ in central PbNe with respect to PbAr collisions. This is qualitatively consistent with 3+1D hydrodynamic predictions including ab-initio descriptions of the nuclear structure. The results provide the first experimental confirmation of the distinctive bowling-pin shape of the ground-state $^{20}$Ne nucleus, validating at the same time the hydrodynamic description of the hot medium formed in high-energy collisions involving light ions.

Résumé : Background and Aims Atezolizumab–Bevacizumab (AtezoBev) was the first immunotherapy approved for hepatocellular carcinoma (HCC) in France, with initial trials primarily involving patients with viral‐related liver disease. This prospective study aimed to evaluate the efficacy of AtezoBev in a French HCC population predominantly affected by non‐viral liver disease. Methods Data from 545 HCC patients treated with AtezoBev as first‐line systemic therapy were collected from 32 French centres in the CHIEF cohort between July 2020 and January 2023. Kaplan–Meier analysis evaluated overall survival (OS) and progression‐free survival (PFS), while log‐rank tests assessed the impact of baseline characteristics. Results Median age was 69, with 81% Child‐Pugh A and 19% Child‐Pugh B. Liver disease was primarily alcohol‐related (30%) or viral (16%), with mixed aetiology with at least alcohol consumption in 58%. At AtezoBev initiation, 72% of cases were treatment‐naive, 31% were BCLC‐B and 64% were BCLC‐C. Median OS was 23.1 months, with a 12‐month survival rate of 66%. OS was higher in BCLC‐B patients (27.8 months) compared to BCLC‐C (17.2 months, p = 0.0043) and in Child‐Pugh A (26.4 months) compared to Child‐Pugh B (10.6 months, p < 0.001). Median PFS was 5.2 months, with BCLC‐B patients showing significantly longer PFS (6.7 months vs. 3.7 months for BCLC‐C, p = 0.05). Conclusion Real‐world data from the CHIEF cohort demonstrate AtezoBev's effectiveness in a large French HCC population, showing survival and response rates comparable to the IMbrave150 study. These findings validate AtezoBev as effective in routine practice across diverse clinical profiles.
