Résumé : Mucopolysaccharidoses are a class of lysosomal storage diseases, characterized by enzymatic deficiency in the degradation of specific glycosaminoglycans (GAG). Pathological accumulation of excess GAG leads to multiple clinical symptoms with systemic character, most severely affecting bones, muscles and connective tissues. Current therapies include periodic intravenous infusion of supplementary recombinant enzyme (Enzyme Replacement Therapy-ERT) or bone marrow transplantation. However, ERT has limited efficacy due to poor penetration in some organs and tissues. Here, we investigated the potential of the β-D-xyloside derivative odiparcil as an oral GAG clearance therapy for Maroteaux-Lamy syndrome (Mucopolysaccharidosis type VI, MPS VI). In vitro, in bovine aortic endothelial cells, odiparcil stimulated the secretion of sulphated GAG into culture media, mainly of chondroitin sulphate (CS) /dermatan sulphate (DS) type. Efficacy of odiparcil in reducing intracellular GAG content was investigated in skin fibroblasts from MPS VI patients where odiparcil was shown to reduce efficiently the accumulation of intracellular CS with an EC50 in the range of 1 μM. In vivo, in wild type rats, after oral administrations, odiparcil was well distributed, achieving μM concentrations in MPS VI disease-relevant tissues and organs (bone, cartilage, heart and cornea). In MPS VI Arylsulphatase B deficient mice (Arsb-), after chronic oral administration, odiparcil consistently stimulated the urinary excretion of sulphated GAG throughout the treatment period and significantly reduced tissue GAG accumulation in liver and kidney. Furthermore, odiparcil diminished the pathological cartilage thickening observed in trachea and femoral growth plates of MPS VI mice. The therapeutic efficacy of odiparcil was similar in models of early (treatment starting in juvenile, 4 weeks old mice) or established disease (treatment starting in adult, 3 months old mice). Our data demonstrate that odiparcil effectively diverts the synthesis of cellular glycosaminoglycans into secreted soluble species and this effect can be used for reducing cellular and tissue GAG accumulation in MPS VI models. Therefore, our data reveal the potential of odiparcil as an oral GAG clearance therapy for MPS VI patients.

Résumé : The effect of amorphous aluminosilicate precursor nanoparticles on the formation the nanosized zeolites with Faujasite (FAU) and Sodalite (SOD) type frameworks is illustrated using a new synthetic strategy to prepare nanosized zeolites with tailored particle size distribution, morphology and structure. This two-steps synthesis procedure includes: (1) the formation of colloidal suspensions followed by separation of the amorphous precursor nanoparticles, (2) their subsequent transformation into nanosized crystals by treatment with alkali suspensions (NaOH) solutions only. The selective transformation of the amorphous nanoparticles into FAU and SOD nanosized crystals is studied at atomistic and microscopic levels using 29Si and 23Na NMR spectroscopy, XRD and N2 physisorption, respectively. The presence of sodalite cages occluding highly mobile sodium in the amorphous nanoparticles is confirmed by 23Na-1H D-HMQC NMR spectroscopy. The subsequent treatment of these amorphous precursor particles with aqueous sodium hydroxide illustrates that the cations are not only charge compensators but also inorganic templates stabilizing sodalite cages during the long-range ordering in the amorphous particles.

Résumé : The Rank metric decoding problem is the main problem considered in cryptography based on codes in the rank metric. Very efficient schemes based on this problem or quasi-cyclic versions of it have been proposed recently, such as those in the submissions ROLLO and RQC currently at the second round of the NIST Post-Quantum Cryptography Standardization Process. While combinatorial attacks on this problem have been extensively studied and seem now well understood, the situation is not as satisfactory for algebraic attacks, for which previous work essentially suggested that they were ineffective for cryptographic parameters. In this paper, starting from Ourivski and Johansson's algebraic modelling of the problem into a system of polynomial equations, we show how to augment this system with easily computed equations so that the augmented system is solved much faster via Groebner bases. This happens because the augmented system has solving degree $r$, $r+1$ or $r+2$ depending on the parameters, where $r$ is the rank weight, which we show by extending results from Verbel et al. (PQCrypto 2019) on systems arising from the MinRank problem; with target rank $r$, Verbel et al. lower the solving degree to $r+2$, and even less for some favorable instances that they call superdetermined. We give complexity bounds for this approach as well as practical timings of an implementation using Magma. This improves upon the previously known complexity estimates for both Groebner basis and (non-quantum) combinatorial approaches, and for example leads to an attack in 200 bits on ROLLO-I-256 whose claimed security was 256 bits.

Résumé : The Absolute Quantum Gravimeter (AQG) is the world's first industrialgravimeter measuring g with laser-cooled atoms [1]. Today, several unitshave already been delivered to end-users.After reviewing the keyprinciples of the AQG, we will discuss the demonstrated measurementperformances of the AQG in terms of sensitivity, stability andrepeatability. In particular, we report on a reproducible sensitivity togravity at a level of 1 ?Gal in various types of environment (1 µGal =1e-8 m/s2 ~ 1e-9 g). We will also present our on-going efforts towardsthe thorough understanding of the uncertainty budget (accuracy) of thesensor. Finally, we will share the experience that we have acquired overthe past years regarding the operability of the AQG, with a specificfocus on the field version of the sensor.This new type of gravimeter ispresently the only technology that allows for continuous drift-freemonitoring of gravity over timescales from a few minutes to severalmonths, which opens new perspectives for the investigation of bothspatial and temporal gravity variations [2]. Résumé : Background: Safety data about rilpivirine use during pregnancy remain scarce, and rilpivirine plasma concentrations are reduced during second/third trimesters, with a potential risk of viral breakthroughs. Thus, French guidelines recommend switching to rilpivirine-free combinations (RFCs) during pregnancy. Objectives: To describe the characteristics of women initiating pregnancy while on rilpivirine and to compare the outcomes for virologically suppressed subjects continuing rilpivirine until delivery versus switching to an RFC. Methods: In the ANRS-EPF French Perinatal cohort, we included women on rilpivirine at conception in 2010-18. Pregnancy outcomes were compared between patients continuing versus interrupting rilpivirine. In women with documented viral suppression (<50 copies/mL) before 14 weeks of gestation (WG) while on rilpivirine, we compared the probability of viral rebound (≥50 copies/mL) during pregnancy between subjects continuing rilpivirine versus those switching to RFC. Results: Among 247 women included, 88.7% had viral suppression at the beginning of pregnancy. Overall, 184 women (74.5%) switched to an RFC (mostly PI/ritonavir-based regimens) at a median gestational age of 8.0 WG. Plasma HIV-1 RNA nearest delivery was <50 copies/mL in 95.6% of women. Among 69 women with documented viral suppression before 14 WG, the risk of viral rebound was higher when switching to RFCs than when continuing rilpivirine (20.0% versus 0.0%, P = 0.046). Delivery outcomes were similar between groups (overall birth defects, 3.8/100 live births; pregnancy losses, 2.0%; preterm deliveries, 10.6%). No HIV transmission occurred. Conclusions: In virologically suppressed women initiating pregnancy, continuing rilpivirine was associated with better virological outcome than changing regimen. We did not observe a higher risk of adverse pregnancy outcomes.

Résumé : Purpose: Psammocarcinoma (PK) is a rare disease of unknown origin. We aimed to report the characteristics, management and survival of patients operated on for PK within the French Network for Rare Peritoneal Malignancies (RENAPE) expert centers.Patients and methods: All consecutive cases of PK operated within all 26 RENAPE centers between 1997 and 2018 were retrospectively analyzed.Results: Twenty-five patients were identified. The median age was 53 years [range 17–78]. None of the patients had extra peritoneal metastases at diagnosis. A median of 6 cycles of carboplatin-based systemic chemotherapy was delivered in 52% preoperatively (n = 13) and 56% postoperatively (n = 14); associated with placlitaxel for 12 patients. All patients were operated on. The median PCI was 23 [0–33]. Eighty-four percent had a complete cytoreductive surgery through digestive (n = 7), spleen (n = 3), pancreas (n = 1) resections and/or multiple peritonectomies (n = 11). Five patients (20%) had intraperitoneal chemotherapy. Morbidity (Dindo-Clavien ≥3) was 12%. No postoperative death occurred. After a median follow-up of 42 months (range [2–194]), the median overall (OS) and progression-free (DFS) survival times were respectively 128 months and 31 months. Eighteen patients recurred (72%), mainly in the peritoneum (n = 16). Four of them (22%) were reoperated. The 5 and 10-year DFS rates were both 20.3%. The 5 and 10-year OS rates were 62% and 51.7%, respectively. A complete cytoreductive surgery was associated with a better OS and DFS in a univariate analysis.Conclusion: Complete cytoreductive surgery is the cornerstone of the PK's management as a primary treatment. Recurrence remains common and new adjuvant strategies seem needed.
