Résumé : The self-assembly of amphiphilic copolymers into well-defined nanoparticles depends on the interplay between polymer composition, solvent exchange kinetics, and processing conditions. In this study, we explore the anionic ring-opening copolymerization of 1,2-epoxybutane and ethoxyethyl glycidyl ether (EEGE) followed by a deprotection step to synthesize poly(butylene oxide)-stat-polyglycidol (PBO-stat-PG) copolymers with tunable amphiphilicity. Reactivity ratio analysis confirmed the formation of a gradient microstructure, with preferential incorporation of EEGE in the early polymerization stages. The copolymers were subsequently processed via bulk and microfluidic-assisted nanoprecipitation, and their self-assembly behavior was systematically investigated. Bulk nanoprecipitation demonstrated that the hydrophilic/hydrophobic balance plays a critical role in controlling nanoparticle formation, stability, and size distribution. Copolymers with 85/15 and 80/20 (BO/G) ratios exhibited the most favorable properties, leading to small, stable, and uniform nanoparticles, whereas excessive G content (≥70%) disrupted self-assembly, inducing irregular morphologies. To enhance nanoparticle homogeneity and minimize aggregation, hydrodynamic-flow-focusing (HFF) microfluidics with a fixed flow rate ratio and controlled mixing conditions were implemented. The highly reproducible microfluidic process enabled finer control over solvent exchange dynamics, leading to smaller and more monodisperse nanoparticles compared to bulk precipitation. The rapid solvent diffusion in the HFF system promoted fast nucleation, effectively reducing polydispersity and ensuring superior colloidal stability. This study establishes a direct correlation between copolymer composition, nanoprecipitation methods, and nanoparticle characteristics, providing a scalable and reproducible strategy for the design of well-defined amphiphilic nanostructures with a controlled hydrophilic/hydrophobic ratio. The optimized PBO-stat-PG nanoparticles present promising potential for biomedical applications, particularly in drug delivery, where controlled self-assembly and tailored hydrophilicity are crucial for performance.

Résumé : Tissue-nonspecific alkaline phosphatase (TNAP) is primarily known for its role in skeletal mineralization, through the hydrolysis of inorganic pyrophosphate (PPi). Here we demonstrate that TNAP-knockout mice exhibit liver steatosis and reduced serum triglyceride levels, mirroring the effects of choline deficiency, which impairs phosphatidylcholine synthesis, an essential component of VLDL. In fasting WT mice, TNAP inhibition via SBI-425 administration decreases choline levels in blood and liver. Incubating mouse or human serum with SBI-425 inhibits the dephosphorylation of phosphocholine and phosphoethanolamine, an alternative substrate for hepatic phosphatidylcholine synthesis. In hepatocytes, TNAP inhibition impedes proliferation when the medium is supplemented with phosphocholine instead of choline. Recombinant TNAP hydrolyzes phosphocholine and phosphoethanolamine with similar efficiency than PPi. X-ray diffraction and cryo-EM identified the residues in TNAP's active site interacting with phosphocholine, PPi and the TNAP inhibitor. In summary, TNAP is the phosphatase enabling cellular choline uptake during fasting, participating in hepatic lipid metabolism.

Résumé : Background: Access to healthcare and socioeconomic deprivation are intricately linked. No studies have been led to measure the effect of healthcare accessibility on mortality in patients with MS so far. The objective was to examine the influence of travel time to the expert MS centre and of the accessibility to primary healthcare services on excess mortality in MS. Methods: A retrospective observational cohort study recruited patients from 18 French MS expert centres, with an onset of MS between 1960 and 2015 and a follow-up of up to 30 years. Primary health facility accessibility was measured by the Spatial aCcessibility multiscAlar index. Specialist care accessibility was measured by road travel time to the expert MS centre. Excess death rates (EDR) and excess hazard ratios were studied using additive excess hazard models with multidimensional penalised splines. Results: The study included 33,697 patients. Patients with relapsing-onset MS (R-MS) with a travel time of 40 min had the lowest EDR (Men: 1.2 deaths per 100 person-years (95%CI [0.8;1.8]), women: 0.8 deaths per 100 person-years 95%CI[0.6;1.2]), lower than patients who lived further from the centre. No effect of primary care access was found for patients with R-MS, and no effect of accessibility to primary or specialised care was found for patients with primary progressive MS. Conclusion: This study reveals the impact of travel time to neurologists on excess mortality in patients with R-MS in France. This distance bias association highlights the importance of preventing a potential selection of patients followed in MS expert centres.

Résumé : One potential solution to diminish the aviation carbon footprint is to blend Sustainable Aviation Fuel (SAF) into jet fuels. However, the thermal stability and oxidation stability of standard jet-fuels may be affected by the addition of SAF, and a better understanding of liquid fuel oxidation kinetics is needed. The development of a kinetically well-defined reactor to study this phenomenon remains a challenge, and it is in this context that a new Si-glass microfluidic reactor with a fluid supply made of inert polyether ether ketone (PEEK) has been developed. The experimental rig PEEK-ACHU (Autoxidation µChip Heated Unit) was used to investigate the oxidation stability of a SAF surrogate, and the results were compared to Induction Period (IP) measures, obtained in a standard PetroOxy device. A mixture of n-decane and isooctane was used as the surrogate SAF. The thermal oxidation stability of the mixtures was studied in the microreactor, working at conditions up to 453 K and 40 bar, with a gas-liquid segmented flow that allows the liquid oxidation to occur in homogeneous, self-agitated, liquid slugs, without oxygen diffusion / depletion kinetic limitations. PEEK-ACHU enables residence times as long as 3.8 hours to be reached on a 4-inch diameter Si/glass wafer, with visual access to the reactor, and a well-defined kinetic study of the liquid fuel autoxidation. The total hydroperoxide (ROOH) concentration was quantified at the exit of the microreactor using a specifically designed HPLC device with a post-column reactor for iodometric titration. For comparison, the same analysis was conducted on the SAF oxidized in the PetroOxy apparatus, with the same residence time. The results show that the microfluidic apparatus is able to perform reliable autoxidation measurements with well-controlled operating parameters. The visual access of the reactor is relevant for further investigations, such as spectroscopic monitoring of deposit formation.

Résumé : Accurate egocentric body-pose estimation is essential for delivering immersive experiences in Virtual, Augmented, and Mixed Reality (VR/AR/MR) applications. A major challenge in this setting arises from the limited field of view provided by head-mounted cameras, which often leads to self-occlusion and poor visibility of certain body joints—particularly in the lower body. Current state-of-the-art methods typically perform pose estimation frame-by-frame in a view-aligned manner. While this is computationally efficient and performs well for visible joints, it struggles to estimate occluded joints due to the lack of contextual information. In this work, we explore the role of temporal information in improving joint estimation. We propose a lightweight model that refines pose predictions by leveraging a large temporal window. Our results show that incorporating such module improves the accuracy of joint estimation without incurring a large computation overhead.

Résumé : While the Segment Anything Model (SAM) has demonstrated remarkable capabilities in general image segmentation, its direct application to medical images faces significant challenges, leading to domain-specific adaptations like MedSAM. In current medical literature, SAM-like models are often evaluated using "simulated" bounding box prompts derived from ground truth annotations, potentially overestimating their real world performance. This paper addresses this underexplored problem by systematically evaluating the influence of bounding box quality on melanoma segmentation performance using the ISIC 2018 dataset (RGB skin image corpus). Experiments were conducted on both the foundational SAM and MedSAM, utilizing a structured protocol that includes ideal bounding boxes, baseline heuristic prompts (full image, most probable location, centered point for SAM), and controlled perturbations. These perturbations comprise five levels of expanding, shrinking, and shifting the ideal bounding box (random disturbance between 5% and 25%). Our results demonstrate a significant dependence of segmentation performance on bounding box quality for both models. Depending on the type of bounding box perturbation, the Dice score decreases from 84% under ideal conditions to 37% with inaccurate bounding boxes, corresponding to a reduction of more than 50%. Notably, shrinking the bounding box has a more detrimental effect than expanding it, with accuracy dropping substantially (e.g., MedSAM's Dice score decreases from 91.5 to 60.9 under a 25% shrinkage). Moreover, medical pre-training significantly improves robustness, as MedSAM outperforms SAM by approximately 7 Dice points under ideal conditions. These findings highlight that although SAM-like models offer promising capabilities, their reliance on high-quality bounding box prompts currently limits their applicability for fully unsupervised segmentation in medical image processing pipelines. This underscores the need for either human intervention or more advanced automated prompting strategies in clinical applications.
