Résumé : Posttranslational modification of histones during the cell cycle is a major process controlling many aspects of cell division. Among the variety of histone modifications, mitotic phosphorylation of histone H3 at serine 10 (H3S10ph) plays a crucial role, particularly in proper chromosome segregation. Here we aimed at precisely quantifying this phosphorylation dynamics during mitosis in plant cells in order to reveal molecular pathways involved in this process. We describe an analysis pipeline based on 3D image analysis that allows to semiautomatically quantify H3S10ph in mitotic Arabidopsis root cells. We also developed a new method for the compensation of signal attenuation in Z, based on measurement of objects of interest themselves. We show that this new attenuation correction method allows significant gains in accuracy and statistical power. Using this pipeline, we were able to reveal small H3S10ph differences between cells treated with hesperadin, an inhibitor of an H3S10ph kinase, or between Arabidopsis mutants affected in PP2A phosphatase activity. This tool opens new avenues to explore such regulatory pathways in plants, using the wealth of genetic materials available in Arabidopsis. It can also be applied to study other histone posttranslational modifications and more generally to any discrete 3D signals.

Résumé : Monitoring aboveground biomass carbon (AGC) stocks and their changes is crucial for understanding the global carbon cycle and the impact of climate change. Among remotely sensed methods, the use of the L-band (1.4 GHz) vegetation optical depth (L-VOD) derived from passive microwave satellite observations, offers rapid updates for timely monitoring of interannual AGC changes. L-VOD is sensitive to changes in the total water content of vegetation, which is determined by both AGC (the biomass of vegetation) and vegetation moisture content. While several methods have been used to understand and correct for the influence of the latter parameter when inferring AGC from L-VOD, there is still a lack of quantified corrections for its impact. Moreover, varying benchmark biomass datasets and fitting functions are currently used for converting L-VOD to AGC, making it difficult to harmonize or compare AGC estimates at regional and global scales. To address these issues, we first corrected the L-VOD time series for changes in the vegetation moisture content and then implemented a systematic global-scale calibration, resulting in annual AGC data set (called INRAE-BORDEAUX AGC, hereafter IB AGC) from 2010 to 2020 at a 25 km resolution. The accuracy assessments showed that IB AGC had a reasonably good spatial agreement with LiDAR referenced AGC data (R 2 = 0.60). Moreover, when aggregated at the national level, IB AGC exhibited stronger consistency with long-term net changes from country-level forest inventory data (R 2 = 0.62) than other mainstream satellite products. It is expected that IB AGC will provide an independent means for better monitoring the global vegetation carbon stocks and their variability in response to climate change.

Above-ground biomass carbon (AGC) in live vegetation is a main land carbon stock and can increase or decrease from environmental changes and direct human activities such as deforestation or forest management 1,2 . The carrier of AGC, above-ground biomass (AGB), has been recognized as an Essential Climate Variable (ECV) by the Global Climate Observing System (GCOS) 3 , serving as a critical input for constraining Earth system models 4,5 and a requisite for countries in their mandated carbon accounting 6-8 . Résumé : Abstract. Information on the spatial–temporal variability of seasonal snow cover duration over long time periods is critical for studying the responses of mountain ecosystems to climate change. However, this information is often lacking due to the sparse distribution of in situ observations or the lack of adequate remote sensing products. Here, we combined snow cover data from 10 different optical platforms, i.e. SPOT (Satellites Pour l'Observation de la Terre) 1–5, Landsat 5–8, and Sentinel-2A and Sentinel-2B, to build a time series of the annual snow melt-out day (SMOD, i.e. the first day of no snow cover) at 20 m resolution across the French Alps and Pyrenees (43 × 103 km2). We evaluated the pixel-wise accuracy of the computed SMOD using in situ snow measurements at 276 stations. We found that the residuals are unbiased (median error of 1 d) despite a dispersion (RMSE of 28 d), which suggests that this dataset can be used to study SMOD trends after spatial aggregation. We found average reductions of 21.4 d (5.78 d per decade) over the French Alps and 16 d (4.33 d per decade) over the Pyrenees over the period 1986–2023. The SMOD reduction is robust and significant in most parts of the French Alps and can reach 1 month above 3000 m. The trends are less consistent and more spatially variable in the Pyrenees. This dataset is available for future studies of mountain ecosystem changes and is updated every year using Sentinel-2 data.

Résumé : Expanding crop diversity is essential to address the imminent challenges of agriculture. This is especially true for organic farming, which relies on locally adapted species and varieties. Recently, participatory research approaches have emerged as effective means to support this endeavour. In this study, we collaborated with several stakeholders in the Lyon region, France, to evaluate three minor species related to common wheat (Triticum aestivum subsp . aestivum): einkorn ( Triticum monococcum subsp . monococcum), emmer (Triticum turgidum subsp . dicoccum) and spelt (Triticum aestivum subsp. spelta L. Thell). First, we assessed the agronomic characteristics of each species, highlighting a distinction of einkorn that was associated with high tillering, high protein content, a long phenological cycle, small kernels and low relative yields. Second, we compared intra-species variabilities, revealing greater variation in emmer and spelt. Lastly, outcomes of the participatory approach, including testing adaptive methods and fostering collective learning, may interest other participatory research groups.

Résumé : Abstract Background hPG 80 (circulating progastrin), initially recognized for its oncogenic properties due to its direct link to the Wnt signaling pathway, is secreted by cancer cells and detectable in the blood of cancer patients. The ONCOPRO centralized case-control study (NCT03787056) was designed to prospectively evaluate the diagnostic utility of hPG 80 in patients with 16 different types of cancer. Methods hPG 80 levels were measured in 421 patients with 16 newly diagnosed cancers (median age 65.6 years old) using the DxPG80.Lab kit (Biodena Care). The diagnostic performance of hPG 80 (the primary endpoint) was assessed by comparing baseline hPG 80 levels in cancer patients with those of 330 asymptomatic aged-matched healthy subjects from the general population. Results Between 2018 and 2022, a total of 506 cancer patients were enrolled in the study, with 421 assessable across 16 distinct cancer cohorts. hPG 80 concentrations were significantly higher in cancer patients compared to the healthy population (median 3.8 [IQR: 1.0-11.1] vs. 1.9 [IQR: 0.6–4.2] pM, P < 0.0001). hPG80 levels were not impacted by renal failure, liver dysfunction, or cancer-associated inflammation. The diagnostic accuracy in cancer patients was ROC AUC 0.63, 95% CI = [0.59–0.67]. The highest diagnostic accuracy was seen in patients with lung cancer (AUC 0.75, 95% CI [0.68–0.82]; specificity = 88% for hPG 80 > 7.73 pM in patients aged > 58 years old) and hepatocellular carcinoma HCC (ROC AUC 0.75, 95% CI [0.66-083]; specificity = 88% for hPG 80 > 7.73 pM in patients aged > 58 years old). Conclusions This large prospective study confirms that cancer patients have significantly higher hPG 80 blood concentration compared to the healthy population. Incorporating this straightforward ELISA assay into screening programs is warranted. Trial registration NCT03787056.

Résumé : Background: Defibrillation testing (DT) remains recommended during subcutaneous implantable cardioverter defibrillator (S-ICD) implantation due to limited supporting evidence. Objectives: The objective of this study was to evaluate the long-term impact of DT during S-ICD implantation. Methods: The HONEST (coHOrte fraNcaise des dEfibrillateurs Sous cuTanés) study is a nationwide, ongoing observational study, including all S-ICD recipients in France (2012-2019). Five-year endpoints were centrally adjudicated, and propensity score-weighted analyses compared outcomes by DT status. Results: Among 4,924 patients, DT was performed in 4,066 (82.6%), decreasing from 85.4% (2012-2014) to 66.9% in 2019 (P < 0.001). Nontested patients were older (51.2 vs 49.6 years; P = 0.007), had lower left ventricular ejection fraction (37.6% vs 43.3%; P < 0.001), and were more frequently implanted for primary prevention (68.0% vs 62.4%; P = 0.002) and structural heart disease (84.9% vs 76.8%; P < 0.001). DT-related complications occurred in 0.1%, including 2 deaths. Failure rate was 1.0%, with 87.8% undergoing corrective reinterventions. Independent predictors of DT failure were elevated shock impedance (≥89 Ω; OR: 4.60; 95% CI: 2.32-9.66; P < 0.001) and obesity (body mass index ≥30 kg/m2; OR: 2.17; 95% CI: 1.01-4.55; P = 0.007). After adjustment, DT omission was not associated with increased risks of overall mortality (HR: 1.17; 95% CI: 0.86-1.61; P = 0.313), cardiovascular mortality (HR: 1.04; 95% CI: 0.70-1.56; P = 0.846), sudden cardiac death (HR: 0.27; 95% CI: 0.04-1.72; P = 0.167), and appropriate (HR: 1.01; 95% CI: 0.78-1.30; P = 0.945) or inappropriate shocks (HR: 0.98; 95% CI: 0.78-1.23; P = 0.865). Combined rates of ineffective shocks or undetected ventricular arrhythmias were similar (0.05 vs 0.06 per 100 person-years). Conclusions: Our findings suggest that DT can be safely omitted in the majority of S-ICD recipients, whereas selective DT may be considered in higher-risk subgroups. Résumé : Introduction: Suicidal behaviors (SB) in bipolar disorder (BD) are major adverse outcomes that may influence disease progression. While staging models exist for psychiatric disorders, none include suicide. This study aims to stratify suicidal risk in BD, propose a staging model for SB, and assess its clinical utility. Methods: Participants from the FondaMental Advanced Center of Expertise for Bipolar Disorder (FACE-BD) cohort were categorised into five stages (St) based on SB: St0 (no suicidal ideation [SI]), St1 (SI but no suicide attempt [SA]), St2a (non-severe/violent SA), St2b (severe /violent SA), and St3 (multiple SAs). Stages were analysed based on demographic, clinical, cognitive, and biological characteristics using logistic regression. Results: Key differences emerged between stages. St1 showed longer untreated illness and higher lability and lower functioning than St0. St2a was linked to anxiety, substance use disorders, and longer disorder duration, while male gender and lithium bitherapy were protective. St2b exhibited shorter untreated illness and higher childhood trauma (CTQ) scores, with male gender and alcohol use as risk factors. St3 was associated with BD-II, alcohol use, longer disorder duration, and more depressive episodes, but less anxiety. No biochemical or cognitive differences were found across stages. The model was significantly associated with SA occurrence (LRT = 28.74, p < 0.0001). Conclusion: This staging model for suicidality in BD provides a multifaceted approach to risk stratification and predictive insights, although further refinement is needed.

Résumé : Autophagy is essential for homeostasis and nutrient recycling. Its activity increases with aging and in response to deficiencies. The effects of defective autophagy on root metabolism have not yet been described. Addressing this question through root proteome analyses, we found that most V-ATPases were less abundant in the roots of autophagy mutants than in wild type. V-ATPases deficit, associated with lower root water contents and lower nitrate, magnesium, and potassium concentrations, indicated that the disturbance of cellular ion and water management in autophagy mutants was likely related to vacuole function. Isotopic δ 13 C analyses and leaf temperature measurements using thermography showed that water deficit in autophagy mutants was not due to excess transpiration, as the conductance of stomata was reduced in mutants compared to wild type. Many proteins related to the catabolism of amino acids and lipids and the tricarboxylic acid (TCA) cycle were over-abundant in atg mutants. The increase in several proteases that paralleled amino acid catabolism suggested that in the absence of autophagic flux, compensatory processes could be established to degrade proteins, recycle amino acids, and fuel TCA. Whether the V-ATPases defect affects energy metabolism and promotes lipid and amino acid catabolism to compensate and fuel TCA remains to be explored. In conclusion, this report establishes for the first time a correlation between autophagy and vacuole function through V-ATPases, particularly with regard to water and ion management. Additionally, this report shows the exacerbation of amino acid catabolism in relation to the stimulation of the TCA cycle in autophagy mutants.
